Objective: The aim of this study was to explore expression of ENO1 in HBV associated hepatocellular carcinoma,to explore the effect and possible mechanism of ENO1 on biological activity of hepatoma cells.Methods: ENO1 expression levels in HCC and non-cancerous liver tissues ?Hep G2 cells and seru m were examined by quantitative real-time reverse transcription PCR(q RT-PCR),Western blot,ELISA respectively.After ENO1-specific si RNA transfection,The effect of ENO1 on the proliferation,migration and invasion of Hep G2 cells were examined by the ex periments of CCK-8,colony formation,cell migration and invasion assays.Notch signaling pathway downstream target genes HEY1,HES1,HES5 were examined by realtime-PCR,NICD and EMT related protein snail?slug??-Catenin?E-Cadherin and Vimentin were examined by Western blot.Results:ENO1 was highly expressed in HBV associated HCC and Hep G2.215 cell lines.The down-regulation of ENO1 significantly reduced the ability of proliferation,migration and invasion in HCC cells.The expression of HEY1,HES1,HES5,NICD,snail and slug was decreased by the down-regulation of ENO1 expression.However,the expression of ?-catenin,E-Cadherin and Vimentin did not change.Conclusion: ENO1 has vital role in the development of HCC and is closely related to HBV infection,ENO1 may effect its biological activity through the Notch signaling pathway.
