ObjectiveTo observe the effect of Jianpi Bushenjiedu Recoction on immune function of mice with Lewis lung cancer and its anti-tumor mechanism.To provide experimental basis for the treatment of non small cell lung cancer.MethodsLewis lung cancer cells were cultured in order to achieve the required number of conventional inoculation in C57BL/6J mice,were randomly divided into normal group,model group,chemotherapy group,Chinese medicine group,combined group.After 14 days of treatment,the blood from the eyeball,were stripped of tumor tissue,spleen,tumor inhibitory rate,spleen index;ELISA method was used to detect the serum levels of IL-2,IFN-?;to observe the tumor ultrastructure by transmission electron microscope;and the loss of content weredetected in spleens of CD4~+CD25~+in mice.Results1.ELISA determination results showed that IL-2 group content than normal group treatment were lower than normal group,Chinese medicine group,although the content is low,the difference was not statistically significant,there was statistically significant difference between the model group and the chemotherapy group content and normal group(P<0.05);the content of the Chinese traditional medicine group and integrated group and model group withhigh content of IL-2.The difference was statistically significant(P<0.05),chemotherapy group compared with the model group,the difference was not statistically significant.Each group ofIFN-? were decreased compared with that in the normal group,the difference was statistically significant(P<0.05);the Chinese medicine group compared with the model group,the difference was statistically significant(P<0.05).2.Transmission electron microscopy results showed that the number of cancer cells in model group,cell size,nuclear membrane integrity,cytoplasmic organelles have abundant cytoplasm,rare cell aggregation;traditional Chinese medicine group became smaller and round,uniform chromatin aggregation;group DDP cells decreased,a cell membrane is not continuous,a large number of cells cytoplasmic margination,nuclear accumulation,a small amount of nuclear damage,no complete structure,fewer organelles,large vacuoles,lysosomes of focal things,there is a small amount of apoptotic bodies;combined group had a large number of apoptosis cells,aggregation of chromatin,the cytoplasm dissolved,organelles decreased obviously,there are many vacuoles,lysosomes karyopyknosis necrosis,aggregateon.3.Flow cytometry results showed that mouse spleen CD4~+CD25~+regulation T cells compared with the normal group content,traditional Chinese medicine group,model group,chemotherapy group was significantly higher,the difference was statistically significant(P<0.05),integrated group ratio is higher than the normal group,but the difference was not statistically significant(P>0.05).Compared with the model group,the chemotherapy group,the traditional Chinese medicine group,the combined group was significantly lower,the difference was statistically significant(P<0.05).Conclusion1.Jianpi Bushenjiedu Recoction can inhibit the growth of Lewis lung cancer mice;2.Jianpi Bushenjiedu Recoction enhance the expression of immune factor IL-2,IFN-?,protects mice immune organs;3.Jianpi Bushenjiedu Recoction can induce apoptosis of tumor cells;4.Jianpi Bushenjiedu Recoction can down regulate CD4~+CD25~+regulatory T cells in spleen of Lewis lung cancer,and play a synergistic and attenuated role in combination with che-motherapeutic drugs.
