Objective To certain the Hui Medicine Smelly shell worm different dose group influence on A549 lung cancer survival in mice and the tumor inhibition as well as the effect of medicated serum with Hui Medicine Smelly shell worm on growth and proliferation of Human Lung Adenocarcinnma A549 Cells.Methods 50 C57BL/6J A549 lung cancer in mice were randomly divided into a tumor-burdened model control group,Cyclophosphamide(CTX)group,low dose Hui medicine Smelly shell worm group,medium dose Hui medicine Smelly shell worm group and high dose Hui medicine Smelly shell worm group.Observed 20 days,recorded of mice general living status;Calculated each tumors in mice weight and inhibitory rate;We used 40 male SD rats were randomly divided into normal saline(NS)group,low dose Hui medicine Smelly shell worm group,medium dose Hui medicine Smelly shell worm group and high dose Hui medicine Smelly shell worm group by weight.By adopting the method of serum pharmacology,dreparation of medicated serum.Observed the concentration of A549 cell morphological changes;determined by methyl thiazolyl tetrazolium(MTT)method is used to detect different concentrations drug-containing serum on A549 cell growth inhibition rate;Flow cytometry to detect different dose of medicated serum on apoptosis of A549 cells.Results(1)The inhibitory of mice.Each tumor had reduced in varying degree after treatment:inhibitory rate for CTX group is 40%,the high dose Hui medicine Smelly shell worm group,inhibitory rate for 30.17%,other medium dose Hui medicine Smelly shell worm group is28.87%,low dose Hui medicine Smelly shell worm group is 26.72%.Compared to the control group,the difference is statistically significant.(P<0.05).(2)Observed different concentrations of serum medicated A549 cell morphological changes under the microscope.Compared with the control group,cell morphology is irregular,the membrane appeared different degree of retraction,nucleus pycnosis,and increased cells spacing in Smelly shell worm groups.(3)Determined by MTT method to detect different doses drug-containing serum on A549 cell growth inhibition rate.Compared with the control group,The experimental group cell inhibition rate is higher than the control group at 72 hours(P<0.05),Hui medicine Smelly shell worm high dose group cell inhibition rate is higher than the low dose group at 72hours(P<0.05),Hui medicine Smelly shell worm groups of 72 hours inhibition rate is higher than 24 hours(P<0.05).(4)Flow cytometry to detect different dose of medicated serum on apoptosis of A549 cells.Hui Medicine Smelly shell worm can induce the aopptosis of A549 cells,the apoptosis rate rises with increasing doses,and time was a positive correlation.The high dose group of Smelly shell on A549 cells for 72 h had the strongest effect,cell apoptosis rate can reach 9.0%.ConclusionsHui Medicine Smelly shell worm has inhibition of the growth of A549 lung cancer,can induced tumor cell apoptosis.
