Expression And Significance Of ITGA5 And Caspase 3 In Hepatocellula Carcinoma

Objective To investigate the expression and clinical significance of integrin ?5(integrin ?5,ITGA5)and caspase 3 in hepatocellular carcinoma and adjacent tissues.Methods 1.Immunohistochemical Elivision two step method was used to detect the expression of ITGA5 and caspase 3 in paraffin sections of 36 cases of hepatocellular carcinoma tissues and paraffin specimens of 23 cases of adjacent tissues.The expression of ITGA5 and caspase 3 in hepatocellular carcinoma and its relationship with clinicopathological features were analyzed.2.The relative expression of ITGA5 and caspase 3 mRNA and protein were detected by fluorescence quantitative RT-PCR and Western blot in 25 patients with hepatocellular carcinoma after surgical resection.3.the experimental data were analyzed by spss20.0 statistical software.The measurement data was expressed with ?_Results 1.Immunohistochemical staining results ±s,the two groups were compared by independent samples t test;Count data was compared by chi square or Fisher's exact test.Correlation analysis of two variables using Spearman correlation analysis.The difference was statistically significant with P<0.05.1.1 ITGA5 protein was mainly expressed in cell membrane and cytoplasm,the positive expression rate in hepatocellular carcinoma tissues and adjacent were 88.89%(32/36)and 43.48%(10/23),respectively,the difference was statistically significant(?2=14.109,P<0.01).The expression of ITGA5 in hepatocellular carcinoma tissues was correlated with TNM stage and metastasis(P<0.05).The positive expression rate of ITGA5 was higher in TNM stage ?~? and metastases of hepatocellular carcinoma.1.2 Caspase 3 protein was mainly expressed in cell cytoplasm,the positive expression rate in hepatocellular carcinoma tissues and adjacent were 30.56%(11/36)and 86.96%(20/23),respectively,with a significant difference(?2=17.903,P<0.01).The expression of caspase 3 in hepatocellular carcinoma tissues was correlated with HBsAg,TNM stage and tumor metastasis(P<0.05).The positive expression rate of caspase 3 was lower in Hepatitis B virus surface antigen positive?TNM stage ?~? and metastasis of hepatocellular carcinoma.2.Fluorescent quantitative RT-PCR showed that the relative expression of ITGA5 mRNA in hepatocellular carcinoma tissues(3.188±1.339)was higher than in the adjacent tissues(0.388± 0.238);the relative expression of caspase 3 mRNA in hepatocellular carcinoma tissues(0.209± 0.108)was lower than in the adjacent tissues(5.755±2.522).The difference was statistically significant(P<0.05).3.Western blot results showed showed that the relative expression of ITGA5 protein in hepatocellular carcinoma tissues(0.583±0.041)was higher than in the adjacent tissues(0.048±0.029);the relative expression of caspase 3 protein in hepatocellular carcinoma tissues(0.168± 0.072)was lower than in adjacent tissues(3.885±0.629).The difference was statistically significant(P<0.05).4.The correIation anaIysis indicated that there was a negative reIationship between expression of ITGA5 and caspase 3 in hepatocellular carcinoma tissues(r =-0.341,P<0.05).Conclusion The expression of ITGA5 was increased in hepatocellular carcinoma andwas negatively correlated with caspase 3,which may be associated with metastasis ofhepatocellular carcinoma.
