Objectives: To determine the type and correlation of susceptibility genes and resistance genes in HLA-DRB1 alleles in patients with Tibetan cystic and alveolar hydatid diseases,and to provide the basis for the genetic characteristics and infection mechanism of Tibetan hydatid diseases.Methods: Patients with Tibetan cystic and alveolar hydatid diseases in Yushu of Qinghai Province and Guoluo Tibetan Autonomous Region,and unrelated healthy people in this area were selected as subjects.The method of case control was used.There were 63 cases of cystic echinococcosis and 73 alveolar echinococcosis in case group,with a total of 136 cases.There were 60 cases in controls group.In hydatid disease designated hospital of province and state,the cases of cystic and alveolar echinococcosis were diagnosed by serological ELISA screening,surgery,B-mode ultrasound or CT.Venous blood samples were collected for 3 to 5mL from patients with AE(alveolar echinococcosis)and CE(cystic echinococcosis)and healthy controls.EDTA-Na2 was used for anticoagulation,then specimens were mixed.DNA was extracted from blood.The polymerase chain reaction-sequence based typing(PCR-SBT)technique was used for genotyping of HLA-DRB1,and the comparison of the gene frequency.The experimental data were analyzed by Hardy-Weinberg(H-W)genetic equilibrium.Fisher exact and χ2 test were used for analysis statistically between cystic/alveolar echinococcosis case group and controls group respectively.The correlation between the disease and the HLA-DRBI allele was expressed by calculating the odds ratio(OR)and 95% confidence(CI)intervals.The SPSS19.0 software was used for statistical analysis.Results 1.Frequency of HLA-DRB1*04 in alveolar echinococcosis case group was low than that in controls group,which was 20.55%,30.00% respectively.The differences in the distribution of allele genotypes were statistically significant(χ2=4.71,P<0.05,OR=0.47).2.Frequency of HLA-DRB1*04 in cystic echinococcosis case group was low than that in controls group,which was 20.63%,30.00% respectively.The differences in the distribution of allele genotypes were statistically significant(χ2=4.31,P<0.05,OR=0.47).3.Compared with cystic/alveolar echinococcosis case group and controls group respectively.There were no found statistical differences of susceptibility genes between them.One case of HLA-DRB1 allele in alveolar echinococcosis group of may contain variation.Conclusions: 1.PCR-SBT technology was firstly applied to research the HLA-DRB1 allele genotyping in the Tibetan population patients with hydatid disease on the Tibet Plateau,which provided technological means for future researches on susceptibility and resistance gene of hydatid disease.2.HLA-DRB1*04 genotypes maybe relate to the resistance of cystic and alveolar echinococcosis in the Tibetan population patients on the Tibet Plateau and its are resistance genes,which also offered theoretical reference to the diagnosis and treatment of hydatid disease.
