Objective: To investigate the effect of hydrogen sulfide to the protein of the ERK?JNK and p38 on the MAPK signaling transduction pathway of burnt rat dermal macrophages cultured in vitro.Method: Subjects were selected for burnt?and treated?rats with dorsal wound dermal macrophages.thirty healthy SD rats?male and female?were selected randomly,five of them are selected for normal feeding,the others are burned and fed.One of the nomal feeding rats was randomly selected as normal control group?Group 1?.Twelve of the burned and fed rats were randomly divided into four groups as the experimental group : Burnt control group?Group 2?,NaHS?exogenous H2S donor?group1h,6h,12h?Group 3?,Glibenclamide group?KATP channel blocker?1h,6h,12h?Group4?,NaHS+Glibenclamide group 1h,6h,12h?Group 5?.the normal control group received no intervention,the dorsum of the rats in rest groups were made into deep II ° 5% TBSA burnt injury?wound pathology confirmed?model groups,to separate macrophages into seventy-eight culture flasks,the normal control group and burnt group were cultured normally,to be stable after being cultured,normal control group and burnt control group were added with 200 ul PBS,with the final concentration of200umol/L NaHS was added to the culture medium in the NaHS group,respectively.1h,6h,12 h,with the final concentration of 200umol/L glibenclamide was added into the culture medium in the GLBN group,respectively1 h,6h,12 h,with the final concentration of 200umol/L NaHS and the final concentration of 200umol/L glibenclamide in the NaHS+GLBN group,respectively 1h,6h,12 h.To analyze the three kinds of protein content by the Western Blot at the same time respectively.Results: To compare the expression of ERK protein at different time points in each group :there was significant difference between normal group and burn group?P <0.05?,the NaHS group had a significant difference compared with each group?P <0.05?,there was significant difference between NaHS+GLBN group and GLBN group?P < 0.05?,there was significant difference in the expression of ERK protein between GLBN group and burn control group?P < 0.05?.To compare the expression of JNK protein at different time points in each group :there was significant difference between normal group and burn group?P <0.05?,the NaHS group had a significant difference compared with each group?P <0.05?,there was significant difference between NaHS+GLBN group and GLBN group?P < 0.05?,there was significant difference in the expression of JNK protein between GLBN group and burn control group?P < 0.05?.To compare the expression of p38 protein at different time points in each group :there was significant difference between normal group and burn group?P <0.05?,the NaHS group had a significant difference compared with each group?P <0.05?,there was significant difference between NaHS+GLBN group and GLBN group?P < 0.05?,There was significant difference in the expression of P38 protein between GLBN group and burn control group?P < 0.05?.Conclusion: The expression of ERK decreased and the expression of JNK and p38 increased under the condition of burn stress after burn;Adding the small amount of H2S after burn,the expression level of ERK protein increased,the relative expression of JNK and p38 protein decreased;Glibenclamide can inhibit the K+ channel,while the expression of ERK protein decreased,the relative expression of JNK and p38 protein increased;Exogenous H2S can increase the expression of ERK and decrease the expression of JNK and p38 on the MAPK signaling pathway in macrophages by K+channel.
