Objective: To establish the burn model(rats),burn skin basal macrophages were extracted,exogenous H2 S was added by Western blotting(Western,blotting,WB)method for the determination of protein content in cultured macrophages in p-ERK,p-p38 and p-JNK,to explore the effects of hydrogen sulfide on cultured skin macrophages p-ERK,p-p38 H2 S and p-JNK,to explore some of the mechanisms involved in wound healing.Method: The research object of this experiment was to burn wound skin base extraction of macrophages.In 30 randomly selected 5 normal feeding,the remaining 25 do burn treatment,from 5 normal rats 1 rats were randomly selected as normal group,25 rats in burned rats randomly selected 12 rabbits(divided into burn group,glibenclamide group,Na HS+GLBN group and Na HS group),3 rats in each group.In the cell culture medium of glibenclamide,the final concentration was added into the cell medium of 200umol/L,it effected of 1 h,6 h,12 h separately;In the cell medium of Na HS + glibenclamide,the concentration of Na HS was 200umol/L,the concentration of Glibenclamide was 200 mg / l,and the concentration of Glibenclamide / l was 200umol/L,and the interaction of 1 h,6 h and 12 h.The protein content of p-PERK?p-p38?p-JNK in macrophages were measured by western blot separately after reaching the time of action.Results:(1)To compare the expression of p-ERK protein at different time points in each group(P < 0.05):there was marked difference between normal group and burn group,the Na HS group had a significant difference compared with each group,there was significant difference between Na HS+GLBN group and GLBN group,there was significant difference in the expression of p-ERK protein between GLBN group and burn control group.(2)To compare the expression of p-JNK protein at different time points in each group(P < 0.05):there was marked difference between normal group and burn group,the Na HS group had a significant difference compared with each group,there was significant difference between Na HS+GLBN group and GLBN group,there was significant difference in the expression of p-JNK protein between GLBN group and burn control group.(3)To compare the expression of p-p38 protein at different time points in each group(P < 0.05):there was marked difference between normal group and burn group,the Na HS group had a significant difference compared with each group,there was significant difference between Na HS+GLBN group and GLBN group,There was significant difference in the expression of p-p38 protein between GLBN group and burn control group.Conclusion: The MAPK pathway of rat skin macrophages in vitro was affected by exogenous hydrogen sulfide,which promoted the expression of p-ERK and inhibited the expression of p-JNK and p-p38.
