Objective: To investigate the SHH signaling pathway in Ptch-1 and Smo protein expression in different grade glioma and normal brain tissue and its correlation.Methods:1 To collect the glioma specimens from 2014 to 2016 which were all from the neurosurgery of our hospital.(On the basis of "Chinese CNS glioma diagnosis and treatment guidelines" in WHO classification of tumors of the central nervous system will be divided into glioma WHO ?-? level,?,? level for low grade glioma,?,? for high grade glioma.So we divided the experiment into high grade glioma group and low grade glioma group).2 Will different grade glioma group 65 cases(41 cases of high grade glioma and 24 cases of low grade glioma)and 10 cases of normal brain tissue which were all fixed by formaldehyde.20 cases of different grade glioma group(including 10 cases of high grade glioma and 10 cases of low grade glioma)and the normal brain tissue 10 cases were all frozen in-80? refrigerator.(the normal brain tissue was all from craniocerebral injury patients;before operation,we had told family members the use of the tissue and got their agreement)3 Using the Immunohistochemistry to detect Ptch-1 and Smo protein expression in each group.4 Using the q RT-PCR to detect Ptch-1 mRNA and Smo mRNA expression in each group.5 Using the Western Blot to detect Ptch-1 and Smo protein expression in each group.6 Using the statistical software IBM SPSS 21.0 to analyze data.Results: 1 Immunohistochemical results: protein positive expression rate of Ptch-1: low grade glioma was 62.5%(15/24);high grade glioma was 92.7%(38/41);the positive rate difference was statistically significant(P<0.05).Ptch-1 in the normal brain tissue protein positive expression rate was 10%(1/10).Between low grade glioma and normal brain tissue and high grade glioma and normal brain tissue the difference had statistical significance(P<0.05).Smo protein positive expression rate: low grade glioma was 75.0%(18/24);high grade glioma was 95.1%(39/41);the positive rate difference was statistically significant(P<0.05).Smo in normal brain tissue protein positive expression rate was 20%(2/10).Between low grade glioma and normal brain tissue and high grade glioma and normal brain tissue the difference had statistical significance(P<0.05).2 qRT-PCR results: the expression of target protein mRNA was found in all groups.The high grade glioma relative expression of Ptch-1 mRNA was 0.0027659±0.0028038;the low grade glioma in the relative expression of Ptch-1 mRNA was 0.0018364±0.0015966.The relative expression quantity difference had no statistical significance(P=0.374>0.05).The Ptch-1 mRNA relative expression in normal brain tissue was 0.0006005±0.0005233.Between low level glioma and normal brain tissue and high grade glioma and normal brain tissue the relative expression difference had statistical significance(P<0.05).The high grade glioma relative expression of Smo mRNA was 0.0078187±0.0054682.The relative expression of low grade glioma in the relative expression of Smo mRNA was 0.0037597±0.0016477.The relative expression difference was statistically significant(P<0.05).The relative expression in normal brain tissue Smo mRNA was 0.0007974±0.0005546.Between low level glioma and normal brain tissue and high grade glioma and normal brain tissue the relative expression difference had statistical significance(P<0.05).3 Western Blot results: the high grade glioma relative expression of Ptch-1 was 0.6967±0.1880;the low grade glioma in the relative expression of Ptch-1 was 0.4771±0.1187.The relative expression quantity difference had statistical significance(P<0.05).The Ptch-1 relative expression in normal brain tissue was 0.3155±0.1314.Between low level glioma and normal brain tissue and high grade glioma and normal brain tissue the relative expression difference had statistical significance(P < 0.05).The high grade glioma relative expression of Smo was 0.6477±0.1730;relative expression of low grade glioma in the relative expression of Smo was 0.4976±0.1039.The relative expression difference was statistically significant(P<0.05).Relative expression in normal brain tissue Smo was 0.2703±0.0914.Between low level glioma and normal brain tissue and high grade glioma and normal brain tissue the relative expression difference had statistical significance(P<0.05).Conclusions:1 Ptch-1 and Smo are expressed in the glioma and normal brain tissue.But the amount of glioma tissue is obviously increased normal brain tissue.SHH signaling pathway in glioma is active,and it may be involved in the occurrence of glioma.2 The expression of Ptch-1 and Smo in high grade glioma significantly higher than the low grade glioma.This prompt that with SHH signaling pathway activated degree increased,glioma malignant degree also increased accordingly.
