Objective: To investigate the inhibiting effect of allicin combined with 5-FU in nude mice xenograft tumor of adenoid cystic carcinoma ACC-M cells,and to offer theoretical basis and experimental evidence on combination therapy of allicin and 5-FU in treatment of the cancer.Methods: 30 Balb/c nude mice weighing(18±2)g and at the age of 5-6 weeks were housed in SPF laboratory for 1 week.The concentration of adenoid cystic carcinoma ACC-M cells was adjusted to 2×107/ml with PBS.0.2ml of which was injected subcutaneously under the right shoulder blade of nude mice to establish xenograft tumor models.24 nude mice with long diameter(a)about 5mm of the tumor were picked from these nude mice after 2 weeks and randomly divided into Experimental Group(Allicin Group,5-FU Group and Allicin combined with 5-FU Group)and Control Group(PSB)with 6 nude mice in each group.The drugs were injected into the abdominal cavity of the nude mice at concentration 20mg/ml by the weight 0.01ml/g,once every two days for three times;The equivalent PBS was injected Control Group(PBS).The xenograft tumor volumes were measured form first day to tenth day,once every two days,a total of six times.The feeding,activity and reaction of nude mice were observed during the experiment process.The growth curves of these tumors were drawn.All nude mice were executed for observing the pathological changing by HE staining after 6 days.Results: 1.The nude mice subcutaneous xenograft tumor model of human adenoid cystic carcinoma ACC-M was successfully established in this experiment.14 days after the inoculation,the tumor lump about 5mm in diameter was in subcutaneous tissue.Xenograft tumor transplantation rate was 100%.2.There was no significant change in the diet and activities of nude mice before drug intervention.The external stimulation of the nude mice in Control Group were significantly lower than those in the Experimental Group till the day when nude mice were sacrificed.And 24 nude mice were alive during the experiment.3.It was shown that the xenograft tumor in Control Group increased significantly while the growth of tumors in experimental groups were lower than those in Control Group,especially in the Allicin combined with 5-FU Group as time went by(p<0.05);There was no significant difference of the average volume of the xenograft tumor before the injection(p > 0.05).After the treatment,the xenograft tumor size in experimental groups were significantly smaller than those in Control Group(p<0.01).The tumor size in5-FU Group were significantly smaller than those in Allicin Group(p<0.01).The tumor size in Allicin combined with 5-FU Group were smaller than those used Allicin or 5-FU alone.4.HE staining: The tumor cells were distributed in denses in Control Group with uniform sizes,high atypia,large nucleus,strong staining and frequent abnormal nuclear division,and no obvious necrosis area.The distribution of tumor cells in allicin group was relatively sparse,and the percentage of necrotic cells in tumor cells was less than10%.There is a large necrotic area in 5-FU group,and the percentage of necrotic cells in tumor cells was about 30%.Fibrous proliferation could be found in allicin combined with5-FU group,some of the tumor cells were swelled and degenerated,and the necrosis of the tumor cells accounted for about 60%.Conclusion: 1.It was a good model establish method with a high rate of tumor transplantation by injecting ACC-M cells subcutaneously.2.Allicin and 5-FU either in separate use or in combination use can effectively inhibit the growth of nude mice xenograft tumor of adenoid cystic carcinoma ACC-M,Pathological examination showed that the tumor tissue of each experimental group had different degrees of pathological changes,especially in the Allicin combined with 5-FU Group.
