Objective: To investigate the effect of micro RNA206(mi R-206)on the expression level of gap junction protein 43 phosphorylation(P-Cx43)in human glioma cell U251.Methods: U251 cells being plated into 6 well culture-plates would be transfected in the condition of synchronization.This experiment was divided into blank control group,negative control group,mi R-206 high expression group and mi R-206 low expression group.No significant sequence plasmid,mi R-206 plasmid and mi R-206-inhibition plasmid were transfected into the negative control group,mi R-206 high expression group and mi R-206 low expression group respectively.The blank control group did not carry out any relevant plasmid transfection.The transfection efficiency was observed under fluorescence microscope after 48 h.The expression level of P-Cx43 in U251 cells was detected by the mean of western blotting.Results: The transfection efficiency of every group was more than 80% under the microscope at 48 h after the termination of transfection.The expression level of P-Cx43 in mi R-206 high expression group was significantly higher than that in blank control group and negative control group,while low expression group was significantly lower than that in blank control group and negative control group(P<0.05).The expression level of P-Cx43 in blank control group was not changed significantly,compared with the negative control group(P>0.05).Conclusion: The blank plasmid,mi R-206 high expression plasmid and mi R-206 low expression plasmid were successfully transfected into human glioma U251 cells by means of chemical transfection reagent,and the model we need could be established.mi R-206 can upregulate the expression level of gap junction protein 43 phosphorylation in U251 cells.
