Chronic alcoholism and binge drinking alcohol cause brain damage.Recently,the studies on exploring the brain damage caused by acute alcohol consumption are mainly to employ biochemical and behavior methods.To investigate the effects of the acute ethanol consumption on the memory of mice,we obtained the neuron signals in hippocampal region of acute ethanol-induced mice using microelectrode array.The experiment using laser speckle contrast imaging system(LSCI)and behavior experiment of T-maze test were designed to verify the accuracy of electrophysiology experiment.The main contents of this paper are as follows: 1.Principle of the electrical signals of brain neurons and the physical characteristics of memory were explored.Then,the applications of neural electrophysiological technique in exploring the effects of acute alcohol on memory of mice were discussed.2.The Matlab codes were programmed,which were used for preprocessing,denoising,feature extraction of spike potential and clustering the neurons in hippocampal CA1 region.3.Designed electrophysiological experiments to obtain the electrical signals of acute alcohol-induced mice(high and low doses)in hippocampal CA1 region and analyzed the change rules of spikes.In order to verify the results of electrophysiological experiment,we measured the local brain blood flow by LSCI and evaluated the spatial ability of ethanol-induced mice using T-maze test.4.Furthermore,experiments were conducted to access the effects of puerarin on discharge rule of acute alcohol-induced neurons and explore the protective effects of puerarin on brain memory.The spikes of acute ethanol-induced mice in hippocampal CA1 regions were obtained by multichannel microelectrode arrays in this study.The results showed that the acute alcohol consumption with high dose decreased the neural firing rate while the acute alcohol consumption with low dose increased the neural firing rate.In addition,the puerarin has opposite results to acute ethanol-induced mice and showed the protective effect on the memory of acute drinking mice.Moreover,the local brain blood flow and behavioral experiments fully demonstrated the validity of the electrophysiological experiment results.In conclusion,the study has a potential for the theoretical and clinical application on exploring the effects of acute alcohol consumption on memory in mice and the protective effects of puerarin on acute alcohol-induced brain damage.
