Increased Expression Of Hsa_circ₀02158 In Bladder Cancer And Its Clinical Significances

ObjectiveThe aim of our study was to discover a certain circ RNA which is closely related to the risks of bladder urothelium carcinoma?BUC?.With the help of literature review and bioinformatics,the association between the target circ RNA and clinical factors would be explored in bladder cancer.Materials and MethodsFollowing informed patient consent and ethical approval of our hospital,total of 60 specimens?tumor tissues and adjacent normal mucosa?were obtained from BUC patients during operation from December 2014 to December 2016.All the specimens were testified by pathologists in our hospital that pure bladder urothelium carcinoma were concluded.With the assistance of literature review and bioinformatics methods,target circ RNA: Hsa_circ₀02158 occurred.Then,specific primer were designed.We made full use of different cell lines including 5637?T24?EJ and SVHUC-1 to verify the quality and availability of the primer and outcome.After that,total RNA extraction and c DNA synthesis were conducted immediately.To validate the existence of Hsa_circ₀02158 in BUC tissues,the product of quantitative real time PCR was sequenced and the sequencing results were aligned with the theoretical sequence.Thus it is of great importance to find the deeper relationship between Hsa_circ₀02158 and BUC.Results1.Compared with NMIBC,the expression of Hsa_circ₀02158 in MIBC was significantly up regulated?4.76±3.192 vs 25.07±23.109?and P value was 0.001.2.Compared with normal samples,the expression of Hsa_circ₀02158 in cancer tissues was significantly up regulated and P <0.053.The relative expression level of Hsa_circ₀02158 in N-patients was lower than that in N+ patients with P=0.001.4.There were no statistical significance between differents groups of clinical factors such as age,gender,status of Lymphovascular Invasion?LVI?and tumor size.Conclusions To sum up,the existence of Hsa_circ₀02158 in the tissues was validated.What'more,Hsa_circ₀02158 was significantly related to the occurrence,progression and migration of BUC,boosting the biological behaviors of the cancer in Chinese population.Our results suggest that Hsa_circ₀02158 may be a new target for treatment of BUC and a potential biomarker in the future.
