Objective To explore the distribution of the C-X-C motif chemokine ligand 12(CXCL12)gene rs1746048 genotype and allele frequency between Intracranial aneurysm(IA)patients and the healthy volunteers in Han Chinese,and evaluate the contribution of CXCL12 rs1746048 to the risk of IA in Han Chinese.Methods A total of 256 unrelated hospital patients were gathered between August 2013 and January 2016 from the Department of Neurosurgery of the Ningbo First Hospital in Ningbo city of Zhejiang province,China.The 361 healthy volunteers were randomly selected from participants in the health examination by Ningbo First Hospital.Both IA and control groups were diagnosed by the imaging examination upon their brains such as CTA?MRA or DSA.The control participants had no symptoms or history of disease,including stroke,autoimmune disease,hematological diseases.Genomic DNA was extracted from peripheral blood samples,using the conventional phenol–chloroform extraction method or the magnetic bead isolation method.The gene polymorphism of CXCL12 rs1746048 were detected by Melting Temperature shift(Tm-shift)polymerase chain reaction(PCR).Statistical analysis were used reveal the correlation between rs1746048 and IA risk in overall study population?female group?male group?the group aged below 65?the group aged 65 or above?female group aged below 65?female group aged 65 or above?male group aged below 65?male group aged 65 or above.Results The genotype of CXCL12 rs1746048 was within Hardy-Weinberg equilibrium law(HWE)(P>0.05).(1)?CXCL12 rs1746048 could be detected in three genotypes: CC?CT?TT.No significant difference was found in the distribution of three genotypes between cases and controls.And no significant difference was found from different groups divided by genders and age respectively(P>0.05);(2)?There was no significant difference in allele frequency of CXCL12 rs1746048 between case and control group ? the group aged below 65 ? the group aged 65 or above?female group aged below 65?male group aged below 65 and male group aged 65 or above(P>0.05);(3)?CXCL12 rs1746048 C allele frequency showed significant association with the risk of IA in the female group aged 65 or above(OR = 2.15,95%CI = 1.13-4.11,P = 0.019).Moreover,rs1746048 was likely to exert its effect under a recessive model in the female group aged 65 or above(OR = 3.77,95%CI = 1.08-13.12,P = 0.03).Conclusions Our results suggested that CXCL12 rs1746048 may contribute to the risk of IA in the female group aged 65 or above and that rs1746048 had a 377% increased risk of IA in subjects aged 65 years or older under a recessive model in Han Chinese.
