Objective The experiment is to investigate the adhesion and proliferation behavior of mouse osteoblasts on cyclic peptide c(RGDfk)-modified titanium surface.Methods Applying the mico-arc oxidation we prepared the porous TiO2 bioceramic film on the surface of titanium plate,and applying coupling effect of polydopamine,the targeted RGD was grafted to the TiO2 bioceramic film.We defined the titanium plate deal with the mico-arc oxidation as MAO group,naming M for short,defined the titanium plate grafted with GRGDSP peptide by coupling effect of polydopamine as linear peptide group,naming X for short,defined the titanium plate grafted with c(RGDfK)by coupling effect of polydopamine as cyclic peptide group,naming H for short.Scanning electron microscopy(SEM)was used to analyze the surface morphology of the three group,energy Dispersive X-ray Detector(EDAX)was used to analyze the surface chemical elements.Through the animal experiments in vitro,we cultivate the MC3T3-E1 cell on titanium plate of all groups.Cell adhesion assay was used to text the optical density(OD)value at 60min?120min?180min?240min,cell proliferation assay was used to text the optical density(OD)value at 1d?3d?5d.Laser confocal microscopy was used to observe the cell morphology at 24 h.Results SEM showed that,the surface morphology of titanium plate dealt with MAO was porous,and the size of the micropore of TiO2 bioceramic film become smaller as the coupling effect of polydopamine,what is more,there were granular RGD peptides in the micropore.EDAX showed that the surface chemical elements on titanium plate of group M was mainly O,Si,P,Ti.on the titanium plate suface of group X and group H,the elements C and N was obviously increased,found that the successful introduction of dopamine and RGD peptides.The results of cell adhesion assay showed that the OD value of group M?X?H was 0.693?0.812?0.867 at the time of 240 min,the result indicated that the number of cell adhere to group H is more than group M?X.The results of cell proliferation assay showed that the OD value of group H was 0.638?0.892?1.193 at the time of 1d?3d?5d,higher than other two group,indicatinng that the cell have more vitality of proliferation on group H.and all the experiments showed significant difference(p < 0.05).Confocal laser experiments showed that the cell on group H had more pseudopodium and wider range of spreading.Conclusions Applying the mico-arc oxidation and coupling effect of polydopamine the functional coating grafted cyclic peptide c(RGDfK)was prepared.Through the animal experiments in vitro,the cyclic peptide c(RGDfK)coating showed good biocompatibility and biological activity,cyclic peptide c(RGDfK)have obvious advantages on Promote cell adhesion and proliferation.
