Objective Observe anti-inflammatory and analgesic effects of Tongduhuoxue decoction,to explore the effects and mechinisms of Tongduhuoxue decoction,which has effects on vertebral artery type of cervical spspondylosis(CSA).Method First choose 200 healthy kunming mices in half respectively male and female,weighing(25±5)g.Randomly divided into 6 groups: normal group,the celecoxib capsules group,the jingfukang group,the high dose group,the middle dose group,the low dose group,10 in each group,By using xylene-induced mouse ear edema test,glacial acetic acid-induced writhing test,hot plate test,evaluation on the anti-inflammation and analgesic action of Tongduhuoxue decoction.Secondly,choose 68 healthy New Zealander rabbits in half respectively male and female,weighing 2.5kg to 3.0kg,were number.choose 10 randomly selected as normal control group,the remaining 68 only for the module,the model group injected hydrocortisone about2.5 mg /100g·d in hind leg rabbit,made kidney Yang deficiency model.The rabbit respectively before and after the building around the TCD detection of vertebral artery and basilar artery blood flow velocity.Select 60 rabbit in building successful,randomly divided into 6 groups:the model group,the celecoxib capsules group,the jingfukang group,the high dose group,the middle dose group,the low dose group.Regularly observe the general situation of the rabbit,measure and record the rabbit body weight changes,hemorrheology,TCD detection,using the ELISA method respectively to detect the contents of cAMP and cGMP in rabbit plasma and ACTH;Serum NO and ET levels;The content of PGE2 in cervical intervertebral disc nucleus pulposus.Take the rabbit brain and cervical intervertebral disc pathological slices,after HE staining under watch pathology morphological structure changes.Result 1.Tongduhuoxue decoction of the experimental research on the anti-inflammatory analgesic action:Compared with normal group,each group had a significantly reduced auricle swelling degree,number of twist body significantly reduced significantly increased,hot plate pain threshold,statistically significant difference(P<0.01 or P<0.05),Which connects high dosel and jingfukang group,celecoxib capsules group anti-inflammatory analgesic action,is no statistical difference between groups(P>0.05).2.Tongduhuoxue decoction of effects and mechinisms on CSA syndrome:Model rabbit combined disease and depression,decreased activity,arch curled up,chills,hair sparse,lose luster,hair loss,reduced the volume of feed intake,drinking water,defecate rare pond,color dark purple tongue,ear,claw performance,such as pathology revealed rabbit brain tissue damage,the organization structure of the cervical intervertebral disc degeneration.Model group rabbit weight loss;Around vertebral artery and basilar artery blood flow velocity slowed;Whole blood viscosity,plasma viscosity increased;cAMP/cGMP ratio decreased;The plasma ACTH and serum NO content;serum levels of ET,higher content of PGE2 in cervical intervertebral disc nucleus pulposus,compared with the normal group had statistically difference(P < 0.01).Each dose group can improve rabbit general situation and different level brain tissue pathological morphology change,intervertebral disc and so on.Compared with model group,the drug group rabbit weight gain;Around vertebral artery and basilar artery blood flow speed;Whole blood viscosity,plasma viscosity decreased;cAMP/cGMP ratio increases;the plasma ACTH and serum NO content increased;Serum ET,reduce content of PGE2 in cervical intervertebral disc nucleus pulposus,but the high-dose group the best,group compared with the other groups have significant difference(P<0.01 or P<0.05).Conclusion 1.Tongduhuoxue decoction has significant anti-inflammatory and analgesic effect and showed some dose-effect relationship,in which the high-dose optimum.2.Tongduhuoxue decoction can improve the rabbit with CSA of shenduxuyu syndrome ships,increasing the weight of the rabbit model,and can improve the rabbit model of the brain stem,pathological cervical disc injury.3.Tongduhuoxue decoction can accelerate the rabbit with CSA of shenduxuyu syndrome artery,basilar artery blood flow velocity,can reduce whole blood,plasma viscosity,improve blood supply of vertebral artery,improve concentration,blood sticky situation.4.Tongduhuoxue decoction can elevated the rabbit with CSA of shenduxuyu syndrome cAMP/cGMP ratio,elevated plasma ACTH content,so as to improve deficiency pathological role.5.Tongduhuoxue decoction can Elevated the rabbit with CSA of shenduxuyu syndrome serum NO,ET level reduced by adjusting the vasoactive factors,improving the state of systolic and diastolic blood vessels.6.Tongduhuoxue decoction can reduce the rabbit with CSA of shenduxuyu syndrome disc nucleus of PGE2 content,reduce the disc inflammatory mediators,delay disc degeneration.The results show that Tongduhuoxue decoction on the CSA of shenduxuyu syndrome rabbit has obvious therapeutic effect between each dose showed some dose-effect relationship,in which the high-dose group optimum.
