ObjectiveTo investigate the effect of Platelet-rich-fibrin(PRF)for wounds with exposed bone.Methods24 male Sprague-Dawley rats(250-300 g,10-12 weeks old)were divided into 3groups(group A,B,C)randomly.A 1.5×1.5-cm full-thickness wound was excised over the posterior scalp area and all the periosteum overlying the skull was removed.The wound was splinted with self-adhesive dressing(DuoDERM ? CGF ?)to stabilize the wound edge.Then wounds of Group A were covered with 1.5ml of PRF which was acquired by centrifugation of blood collected in heart puncture,while those of Group B were covered with the equivalent of fat particles which were grafted from groin of rats,and Group C were covered with the equivalent of normal saline(NS).Then wounds of the three groups were covered with an adhesive dressing Tegaderm?as a seal.Wounds were redressed under general anesthesia on the fourth,eighth and eleventh day.Granulation growth and bone exposed area were quantified by Image J software.ResultsOn the fourth day,the percentage of bone exposure in wounds of the fat-transplantation-group was the lowest among the three groups.On the seventh and eleventh day,all wounds in group A were completely covered by granulation.While there were bones remained exposed in group B and C.Wounds in group A showed accelerated healing significantly comparing with group B and C.Histomorphometry showed that evident lumen structure appeared in all of the three groups.Compared with the control group,transit of erythrocytes were only found in fat-transplantation-group and PRF group,indicating that their blood vessels are moremature.Moreover,the direction of fiber arrangement showed aligned in PRF group,while it's irregular in fat-transplantation-group and control group.at each time point,the bone-exposed area of PRF group was significantly lower than that of the control group.(P<0.01)ConclusionsPRF as a convenient and cost-effective therapy can accelerate wounds healing in bone exposure,shorten the period of treatment and reduce the burden of surgery.
