Objective:1.The purpose of the study is to investigate the expression of Bromodomain protein 4(BRD4)in gastric carcinoma.2.To analyze the relationship between the expression level of BRD4 and pathological features,such as gender,age,smoking and drinking history,postoperative pathological pT staging,histological differentiation,lymph node metastasis,distant metastasis,EGFR level,VEGF level,tumor location,patient survival prognosis.Methods:1.Retrospective analysis of 95 patients who were diagnosed with gastric carcinoma(GC)in the department of general surgery in our hospital(Fuzhou General Hospital)from January 2009 to December 2010.All the patients were carried the gastric cancer radical surgery and used telephone to realize the follow-up survival.The deadline for the follow up was December 15,2015.2.The expression of BRD4 in gastric carcinoma was detected by immunohistochemistry.The data were analysed by SPSS18.0 software package,the relationship between the expression of BRD4 and the clinical pathological features and prognosis in patients with gastric cancer were analysed by chi-square test.3.The single factor analysis and COX proportional hazard regression model were used to analyze the relationship between BRD4 and age,gender,smoking and drinking history,tumor location,histological differentiation,lymph node metastasis,distant metastasis,prognosis,recurrence and clinical pathological features.The analysis of independent risk factors was performed by Kaplan-Meier survival curves.Results:1.Immunohistochemical staining indicated that BRD4 protein mainly accumulated in the nuclei of GC cells.Immunohistochemical analysis revealed BRD4 was overexpressed in GC tissue compared with adjacent normal tissue.2.A total of 95 patients who underwent radical gastrectomy for gastric cancer were enrolled in the study.They were all independently diagnosed with gastric cancer by two pathologists.There were 56 patients who were over than 60 years and accounted for 58.9%,39 patients who were less than 60 years and accounted for 41.1%.Of all the cases,men accounted for 73.7%,women accounted for 26.3%.The pathologic differentiation degree with high,medium and low differentiation accounted for 8.4%,49.5% and 42.1% respectively.In the pT stage,there were 20 people in T1 stage,20 people in T2 stage,21 people in T3 and 34 people in T4 stage.64.2% of the cases had lymph node metastasis.28.4% of the patients had distant metastasis.There were 41.1% of the patients who were still alive at the end of the follow-up.High expression of BRD4 was related to drinking or smoking history(p=0.040),pT stage(p=0.004),lymph node metastasis(p<0.01),VEGF(p=0.007),distant metastasis(p<0.01)and survival(p<0.01).Conclusion:In conclusion,BRD4 was overexpressed in GC,higher BRD4 expression predicted worse prognosis,BRD4 could act as a potential biomarker for prognostic assessment of GC.
