Part1 Detection of TREM-1 expression in gingival tissues of patients with periodontal diseasesObjective To observe the distribution and expression of triggering receptor expressed on myeloid cells-1(TREM-1)in the gingival tissue of patients with different types of periodontitis and to preliminary explore the possible effects of TREM-1 on the occurrence and development of periodontitis.Methods A total of 30 volunteers,including 10 healthy controls,10 patients with gingivitis and 10 cases of chronic periodontitis were involved in this study.The gingival specimens were taken and fixed in 10% buffered formalin for more than 48 h,stained with hematoxylinand eosin for the observation of histological changes under optical microscope,and stained with immunofluorescence for identification of TREM-1 in gingival tissues.Results(1)TREM-1 expressed in the gingival epithelium and connective tissue full layer.(2)TREM-1 fluorescence was observed localized in the cytomembrane and cytoplasm,but not in the nucleus.(3)The expressions of TREM-1 in the health group are mainly distributed in the gingival epithelium,especially in the basal layer,and the specific staining intensity of the gingival tissue was not obvious.(4)Compared with healthy control group TREM-1 positive cells increasedinthe gingivitis group,and the specific staining intensity of gingival tissue was moderately positive.(5)The expression of TREM-1 in the chronic periodontitis groupwas significantly higher than that in healthy control group and the gingivitis group,the number of TREM-1 positive cells increased.Conclusion The expression of TREM-1 in gingival tissues is increased with the severity of chronic periodontitis.This results suggest that TREM-1 as a biomarker might play an important role in human chronic periodontitis.Part2 Detection of s TREM-1in gingival crevicular fluid of patientswith periodontal diseasesObjective To investigate the levels of s TREM-1 in the gingival crevicularfluid(GCF)of individuals without periodontitis and with chronic gingivitis or chronic periodontitis and explore the role of s TREM-1 in the development and progression of chronic periodontitis.Methods A total of 85 volunteers,including 18 healthy controls,18 patients with gingivitis,14 patients with mild chronic periodontitis,15 patients with moderate chronic periodontitis and 20 patients with severe chronic periodontitiswere involved in this study.The concentrations of s TREM-1 in GCF with different types of periodontitis weredetected by enzyme-linked immunosorbent assay(ELISA).The correlation between s TREM-1 concentrations and gingival index(GI),pocket depth(PD)and loss of attachment(CAL)was analyzed.Results The concentrations of s TREM-1 in GCF in chronic gingivitis group,and mild,moderate and severe chronic periodontitis group were significantly higher than those in healthy group(P<0.05).The concentrations of s TREM-1 in GCF in moderate and severe chronic periodontitis group were significantly higher than those in chronic gingivitis group(P <0.05).There was no significant difference in the concentration of s TREM-1 in GCF between mild chronic periodontitis group and chronic gingivitis group(P> 0.05).There was no significant difference in the concentration of s TREM-1 in GCF between moderate chronic periodontitis group and mild chronic periodontitis group(P>0.05).The concentrations of s TREM-1 in GCF in severe chronic periodontitis group were significantly higher than those in mild chronic periodontitis group(P <0.05).There was no significant difference in the concentration of s TREM-1 in GCF between moderate chronic periodontitis group and severe periodontitis group(P >0.05).In addition,there was a significant positive correlation between concentration of s TREM-1 and GI,PD and CAL.ConclusionThe concentrationof sTREM-1 in GCF is closely related to the inflammation of periodontal tissue,which increases with the severity of periodontal tissue inflammation.The detection of s TREM-1 in GCF has potential application value in judging the inflammation of periodontal tissue.
