Objective To observe and explore the effect of TNF-? on cognitive functionin the aged diabetic rats and the central modulating mechanism.Methods Sixty male SD rats were randomly divided into three groups: normal control group,diabetic control group and diabetic inflammation group.The diabetes mellitus rat model was established by glucose-rich and fat-rich diet combined with streptozocin induced by intraperitoneal injection.The experimental arrangement: Normal control group and diabetes control group rats were injected sterile water through tail vein,for the diabetic inflammation group,we used TNF-? 0.4ug/kg,and then the reasults were measured about Morris results,the expression of TNF-?,A? in the hippocampus,the moisture content of brain tissue,the concentration of S100? protein and NSE of correlation index of brain tissue injury in serum on the day before injection,the first,the third,the seventh day after injection.At the same time,the changes of the blood brain barrier structure and function changes were measured.Results Respectively compared with the normal control group and the diabetic control group,the diabetic inflammation group showed a significant decrease result of Morris water maze on the first day after injection and reached the lowest,then increased gradually,on the third day after injection were still significant differences(P<0.05);The expression of TNF-?,A? in hippocampus and the moisture content of brain tissue were significantly increased on the first day after injection and reached the peak(P<0.05)and then decreased gradually,there was significant difference on the third day after injection(P<0.05);The concentration of S100? protein and NSE of correlation index of brain tissue injury in serum were significantly higher and reached the peak(P<0.05),and then decreased gradually,there was significant difference on the third day after injection(P<0.05);The permeability of the blood brain barrier of the elderly diabetic rats were significantly increased on the first day after injection and reached the peak(P<0.05),and then decreased gradually,there were significant difference on the third day after injection(P<0.05).Conclusion 0.4ug/kg TNF-? caused the destruction and permeability increased of blood-brain barrier structure in aged diabetic rats through tail vein injection,and then caused the hippocampus tissueinflammatory injury that may be one of the mechanisms of cognitive function impairment in short time(3 days).Objective To explore the effect mechanism of TNF-? monoclonal antibodyon the TNF-? caused the cognitive dysfunction in aged diabetic rats.Methods Sixty male SD rats were randomly divided into three groups: diabetic group(TT group),diabetic monoclonal antibody group(TA group),diabetic inflammation + monoclonal antibody group(TTA group).The diabetes mellitus rat model was established by glucose-rich and fat-rich diet combined with streptozocin induced by intraperitoneal injection.TNF-? 0.4ug/kg was injected through tail vein in TT group,TNF-? monoclonal antibody 1mg/kg in TA group,for the TTA group,we injected TNF-? monoclonal antibody 1mg/kg,after two hours,TNF-? 0.4ug/kg was injected through tail vein.Then the reasults were measured about Morris results,the expression of TNF-?,A? in the hippocampus,the moisture content of brain tissue,the concentration of S100? protein and NSE of correlation index of brain tissue injury in serum on the day before injection,the first,the third,the seventh day after injection.At the same time,the changes of the blood brain barrier structure and function were measured.Results Compared with TT group,TTA group was higher about the Morris water maze results on the first day after injection,that was significant difference between the two groups(P<0.05);The expression of TNF-?,A? and the moisture content of brain tissue in hippocampus were significantly decreased on the first day after injection,that was significant difference between the two groups(P<0.05);The concentration of S100? protein and NSE of correlation index of brain tissue injury in serum were significantly decreased on the first day after injection,that was significant difference between the two groups(P<0.05);The destruction and permeability of the blood brain barrier were normal on the first day after injection in TTA group,that was significantdifference between the two groups(P<0.05).Conclusion The pre-injection of TNF-? monoclonal antibody through the tail vein could reduce the damage of the blood-brain barrier and the hippocampus inflammatory,that could reduce the damage of TNF-a on the cognitive dysfunction in aged diabetic rats.
