Background Lung cancer is the world's highest mortality of malignant tumors,a serious threat to human life and health.Lung cancer is generally classified into small cell lung cancer(SCL)and non-small cell lung cancer(NSCLC),in which,they account for about 15%and 85% of the total of lung cancer,respectively[1].So,non-small cell lung cancer is the main pathological type of lung cancer.Non-small cell lung cancer is divided into two types,that is,non-squamous cell carcinoma including lung adenocarcinoma,large cell carcinoma and other types,and squamous cell carcinoma(i.e.,epidermoid carcinoma).As with other malignancies,the primary reasons of the occurrence and development of lung cancer include mutations or loss of activity in tumor suppressor genes,as well as oncogene activation.In addition,some biological markers also have a significant impact on the prognosis of patients with lung cancer and the sensitivity to the corresponding drugs[2-5].Therefore,research of molecular phenotype of lung cancer is more beneficial to determining of the phenotype,thereby,providing effective reference indexes for developing individualized treatment regimens and evaluating the prognosis in different patients.Although over the past decade or so,researchers have made some progress in molecular and molecular networks for the development of lung cancer at the protein and gene levels,it has played a huge role in promoting the development and development of new drugs and the search for new lung cancer treatment,But the overall 5-year survival rate in patients with lung cancer has not been improved[6-7].The occurrence and development of malignant tumors are often multi-stage complex processes involving a variety of factors in vivo and in vitro,including multiple genes and signal transduction pathways.In recent years,most of the studies focused on lung cancer-related genes in the process of malignant tumor process,so in-depth study of abnormal changes in lung cancer is the basis for further clarification of lung cancer mechanism.Kinesin is a molecular motor protein based on microtubules,It plays an important role in vesicles,organelles,chromosomes,protein complexes in the transport process,but also with the intracellular material transport and cell decomposition Physiological processes are closely related.Kif18 A,a member of the Kinesin family,is capable of binding to microtubules and thus plays an important role in the localization of membrane organelles and protein complexes by mobilizing energy released by the hydrolysis of ATP along the microtubules[8-12].Studies have shown that,Kif18 A is associated with development and progression of colorectal cancer,breast cancer,liver cancer and gastric cancer[13-19],but not yet for its non-small cell lung cancer malignant process correlation.Objective To understand the expression of Kif18 A in NSCLC tissues,explore the correlation between Kif18 A and clinicopathological data,and study the feasibility of Kif18 A as NSCLC marker,target and prognostic index,providing a new direction for the diagnosis and treatment of NSCLC.Method: 100 NSCLC patients collected in Quanzhou First Hospital of Fujian Affiliated to Fujian Medical University from September 2016 to December 2016 were chosen,and the tissues were collected from 50 cases of adenocarcinoma,50 cases of squamous carcinoma,and 50 cases of the corresponding para-carcinoma tissues each.The clinical data were summarized in the following table.The expression of Kif18 A protein in the cancerous tissues and the adjacent normal tissues of the above-mentioned pathological samples were determined by Western blot and immunohistochemical method,and the correlation between it and the parameters(i.e.,age,sex,pathological type,presence or absence of lymphatic metastasis and TNM stage in lung adenocarcinoma and lung squamous carcinoma patients)were analyzed.ResultsWestern blot analysis showed that the expression of Kif18 A protein in lung adenocarcinoma and lung squamous cell carcinoma was significantly higher than that in adjacent normal tissue(P <0.05).The immunohistochemical results of Kif18 A protein in lung adenocarcinoma and squamous cell carcinoma of lung were significantly higher than those in adjacent normal tissues(P <0.01).According to the correlation between the expression of Kif18 A and clinical pathological parameters,there was no statistically significant difference between male and female patients(P > 0.05),as well as between patients aged > 62(lung adenocarcinoma)/61(squamous cell lung carcinoma)and those aged < 62(lung adenocarcinoma)/61(squamous cell lung carcinoma)(P > 0.05).Moreover,no statistical difference was found between lung adenocarcinoma patients and squamous cell lung carcinoma patients(P > 0.05).Meanwhile,the expression of Kif18 A protein was significantly higher in well-differentiated lung cancer patients than moderately and poorly differentiated ones(P <0.05).Also,it was higher in patients with lymph node metastasis and stage III NSCLC patients,compared with those without lymph node metastasis and stage I + II NSCLC ones,respectively(P <0.05).Conclusion The expression of Kif18 A in NSCLC tissues was higher than that in the adjacent normal tissues.It was associated with the degree of tumor differentiation,presence or absence of lymph node metastasis and TNM staging,but not with patient's sex,age and pathological type.A significantly high expression rate of Kifl8 A was shown by the immunohistochemical method relative to western blot method(P<0.05).
