Objective To observe the intraperitoneal injection of different doses of dexmedetomidine on the expression of Th17/Th22 cells and cytokines in septic mice,and to analyze the effect on liver injury in mice with sepsis,so as to explore the possibility of dexmedetomidine participated in repairing sepsis induced liver injury by adjust the balance of Th17/Th22 cells,and thereby to provide new ideas and methods for the treatment of liver injury induced by sepsis.Methods A total of 90 healthy C57BL/6 mice were selected for the experiment.The mice were divided into six groups according to random number table(n=15): sham-operation group(group S),sepsis control group(group D),15?g·kg-1 exmedetomidine group(group Y1),30?g·kg-1 exmedetomidine group(group Y2),60?g·kg-1 exmedetomidine group(group Y3),15?g·kg-1 exmedetomidine + 1.5mg·kg-1 yohimbine group(group Y4).Group S only opened the abdominal cavity and flipped the cecum without cecal ligation,cecal ligation and puncture(CLP)were conducted in group D,group Y1,group Y2,group Y3 and group Y4 to establish mice model of sepsis induced liver injury,corresponding dose of dexmedetomidine was slowly injected by intraperitoneal injection in group Y1,group Y2,group Y3 about 0.5h after CLP,15?g·kg-1 exmedetomidine + 1.5mg·kg-1 yohimbine was injected intraperitoneally and slowly in group Y4,while mice in group S and group D were injected with equal volume of saline for more than 10 min.Blood samples were taken from the tail vein at 12 h after operation,mice were then put to death by decapitation,liver pathological specimens were obtained quickly.The expression of Th17/Th22 cells in peripheral blood was detected by flow cytometry;the levels of IL-6,IL17 and L22 in peripheral blood were detected by ELISA;ALT and AST levels were detected by automatic biochemical analyzer;pathological changes in the liver tissues were observed by HE staining at the same time.Statistical methods: data were analyzed by SPSS17.0 software package.Results(1)Th17 and Th22 cells were increased in group D when compared to group S;the expression levels of IL-17 and IL-22 were increased sharply;blood ALT and AST values were increased significantly,Th17 and Th22 were positively correlated with ALT and ALT.(2)The frequency of Th17 cells and the level of IL-17 were decreased in peripheral blood of group Y1,group Y2 and group Y3 than those of group D;the frequency of Th22 cells and the secretion of IL-22 were found to be decreased gradually;the amount of IL-6 secretion in blood,as well as liver function indexes of ALT and AST levels were detected to be decreased correspondingly.(3)Frequencies of Th17 and Th22 cells were increased in group Y4 than in group Y1;expression levels of IL-17 and IL-22 were increased;ALT and AST values were increased in blood.However,there was no statistical difference between group Y4 and group D.Conclusion(1)The expression of Th17 and Th22 cells was significantly increased in mice with sepsis,inflammatory factors of IL-6,IL17 and IL22 were significantly increased in blood.(2)Th17/Th22 and IL-17/IL-22 were positively correlated with sepsis induced liver injury indexes of ALT and AST,suggesting that Th17/Th22 might play an important role in the development of sepsis induced liver injury.(3)Dexmedetomidine could down-regulate Th17/Th22 secreting inflammatory cytokines of IL-17/IL-22 in the repairing process of sepsis induced liver injury.(4)Such effect of dexmedetomidine could be antagonized by ?2 AR blocker(aphrodine).
