Objective: This study aims to respectively investigate the RACK1 expression and the infectious status of HR-HPV in normal cervix,precancerous lesion and cervical carcinoma,and to analyse the relationship between the RACK1 expression and the infections status of HR-HPV.Methods: Randomly collected cervical tissue specimens that inspected or treated in our institution from October 2015 to October 2016,15 cases of normal cervix,15 cases of early stage CSCC and 30 cases of CIN I,30 cases of CIN II and 30 cases of CIN III were included.The paraffin slices of all of the specimens and the fresh-frozen tissues of normal cervix specimens and the early stage CSCC were available.Immunohistochemistry analysis and Western-blot assay were performed to investigate the expression of RACK1 in above specimens.Meanwhile,the cervical secretion of all included patients received the diversion hybrid gene chip method to detect the infectious status of high-risk human HR-HPV in these patients.Spearman rank correlation analysis was performed to indicate the relationship between the RACK1 expression and the infections status of HR-HPV.Results:1.Western blot assay revealed that expression of RACK1 protein in early stage CSCC tissues was significantly higher than normal cervix tissues.Immunohistochemistry examination revealed that the positive expression rate of proteins RACK1 in normal cervical tissues,CINI tissues,CINII tissues,CINIII tissues and CSCC tissues gradually increased.The expression of RACK1 in each groups was 21.74%(5/23),27.27%(6/22),63.41%(26/41)and 66.67%(42/63)and was statistically different between each group(P<0.05).2.As the level of cervical lesions deteriorated,the infectious rate of HR-HPV was higher gradually and was statistically different between each group(P<0.05).Meanwhile,the infectious rate of HR-HPV was positively correlation with the expression rate of RACK1 and the correlation coefficient(rs)was 0.266Conclusion: 1.As the level of cervical lesions deteriorated,the expression rate of RACK1 gradually increased and the infectious rate of HR-HPV also exhibited a similar variety trend.2.The expression of the RACK1 showed a positive correlation with the infectious rate of HR-HPV,which may indicate that the expression of RACK1 plays an important role in the HPV mechanism of carcinogenesis...
