Objective:To explore changs of IL-2 and IL-10 in children with acute leukemia(AL)before chemotherapy and after complete response.Methods:Collected 43 pediatric patients who had been respectly diagnosed with ALL(30)and AML(13),and they were in complete relief after chemotherapy,and 20 normal childrens.The concentrations of IL-2 and IL-10 coming from the kids in control group and the patients at preliminary diagnosis and after complete remission were test by flow cytometry,comparing changs of levels of the two cytokines between ALLand AML,or in the three groups.Results: 1.ALL groupPatients at initial diagnosis(3.59±1.18)pg/ml contained much lower IL-2 content than that in healthy childrens(6.22±2.03)pg/ml(P<0.001);Patients at initial diagnosis(15.88±4.73)pg/ml contained much higher IL-10 content than that in healthy childrens(5.69±3.44)pg/ml(P<0.001).Along with the chemotherapy,the levels of IL-2(4.78±1.57)pg/ml obviously increased(P=0.005)while those of IL-10(8.14±2.82)pg/ml declined significantly(P<0.001),but the concents of both of them were not up to the standards of normal kids(P = 0.034 ? P=0.036).there were significant differences among the groups(P <0.05).2.AML groupPatients at initial diagnosis(2.43±1.61)pg/ml contained much lower IL-2 content than that in healthy childrens(6.22±2.03)pg/ml(P<0.001);Patients at initial diagnosis(35.21±13.19)pg/ml contained much higher IL-10 content than that in healthy childrens(5.69±3.44)pg/ml(P<0.001).Along with the chemotherapy,the levels of IL-2(4.24±1.93)pg/ml obviously increased(P=0.048)while those of IL-10(12.06±3.81)pg/ml declined significantly(P<0.001),but the concents of both of them were not up to the standards of normal kids(P = 0.026 ? P<0.001).there were significant differences among the groups(P <0.05).3.Comparison of ALL group and AML groupThe levels of IL-2 of patients who had been diagnosed with ALL were similar with patients who had been diagnosed with AML at initial diagnosis or after complete remission(P =0.165 ? P=0.932),and there were not significant differences among the groups(P >0.05).But the leves of IL-10 of patients who had been diagnosed with AML were much higher than them who had been diagnosed with ALL at initial diagnosis or after complete remission(P=0.001 ? P=0.022),and there were significant differences among the groups(P <0.05).Conclusion:The IL-2 and IL-10 level in children with AL are vital to the disease onset and progression.The IL-2 and IL-10 level can affect the immume function of the children with AL,and the two cytokines can be used as index condition assessment.The high level of IL-10 in children with AML is closely related to its bad prognosis.
