Objective: To investigate the effect and mechanism of CX3CR1 deficiency regulates the hepatocellular carcinoma.and its microenvironment.Methods: In vitro,the THP-1 was treated by 50ng/ml PMA for 48 h,then the THP-1 were became to macrophages(M0).Macrophages were transfected with the Si CX3CR1 RNA targeting and silencing CX3CR1 gene.Then the expression of CX3CR1 in macrophages were detected by reverse transcription PCR(RT-PCR)and Western blotting.The marker of macrophages were detected by RT-PCR and flow cytometry.The Hep G2 and 7721 cells were co-cultured with macrophages or treated by conditional medium,the proliferation,apoptosis,migration and invasion of Hep G2 and 7721 cells were detected by MTT,flow cytometry,wound-healing and Transwell assay,respectively.The expression of p-ERK1/2 which was related to ERK-MAPK signal pathway and p-AKT which was related to PI3K-AKT signal pathway were detected by Western blotting.Results: After si CX3CR1 was transfected in into macrophages,the expression of CX3CR1 in macrophages was significantly inhibited.The marker of M2 macrophages was decreased and the marker of M1 macrophages was elevated,then the proliferation,migration and invasion of Hep G2 and 7721 cells were inhibited when these cells were treated by si CX3CR1 macrophages conditional medium.Conclusion: The deficiency expression of CX3CR1 inhibited HCC progression via enhancing M1-type polarization of macrophages.
