Cervical cancer is a common female-specific cancer worldwide and cause tremendous damage to women,the morbidity and mortality rate of cervical cancer have increased significantly every year.Cervical cancer is the fourth-most common cause of cancer and deaths of cancer in women worldwide.Although great efforts have been made on HPV vaccines to protect women from cervical cancer,it is still the second-most common cause of female-specific cancer after breast cancer.BCL11A(B-cell CLL/lymphoma 11 A,BCL11A),its homologous gene in murine is called Evi9,was first discovered as a common retroviral integration in myeloid leukemia tumors in the BXH-2 mouse and was regarded as a potential oncogene which interacts with BCL6[1].BCL11 A plays an important role in tumors of the hematopoietic and lymphoid tissues,also be referred to as liquid tumors,such as the leukemias and the lymphomas)[2,6].Recent researches showed that BCL11 A also played a critical role in solid tumors,for instance,in the triple-negative breast cancer and non-small cell lung cancer[3,4].Nonetheless,the function of BCL11 A in cervical cancer have not been reported yet.In this study,we explore the expression of BCL11 A in cervical cancer and its underlying mechanisms.PART 1:THE EXPRESSION OF BCL11 A IN CERVICALCANCER TISSUES AND CLINICAL SIGNIFICANCESObjective: To investigate the expression of BCL11 A in cervical cancer tissues and adjacent non-cancerous tissues,then analyze the clinical significance underlying the differential expression of BCL11 A.Methods: Use bioinformatics to investigate the differential expression of BCL11 A in variant cancer tissues and paired non-cancerous tissues.Clinical specimens,including cervical cancer tissues and paired adjacent non-cancerous tissues,were obtained from 62 patients.Evaluate the differential expression of BCL11 A in paraffin embedded and sliced tissues via immunohistochemistry,combined with individual patient data,analyze the clinical significance underlying the differential expression of BCL11 A.Result: BCL11 A expression was detected in all 62 tissues,high expression was found in 50 of 62 cervical cancer tissues(80.6%)and 13 of 62 adjacent non-cancerous tissues(21%),differential expression existed in cervical cancer(P<0.01).No correlations between BCL11 A expression and age(P=0.436)or tumor size(P=0.734)or amounts of lymphocyte(P=0.439)were found.However,patients with lympho node metastasis showed high expression of BCL11A(P<0.05).Patients with high expression of BCL11 A showed better overall survival.BCL11 A expression was mainly located in cell nucleus in cervical cancer tissues and in cytoplasm in adjacent non-cancerous tissues.Conclusion: BCL11 A expression levels were significantly increased in cervical cancer tissues.BCL11 A expression was related to lympho node metastasis,patients with lympho node involvement showed high BCL11 A expression,the overall survival of patients with high BCL11 A expression was better than patients with low BCL11 A expression,but BCL11 A expression couldn't be independent prognostic indicator of cervical cancer.BCL11 A expression was mainly located in cell nucleus in cervical cancer tissues and in cytoplasm in adjacent non-cancerous tissues.PART 2:EFFECTS OF INTERFERENCE OF BCL11AEXPRESSION ON BIOLOGICAL BEHAVIOR OFCERVICAL CANCER CELLS.Objective: Investigated the effect of BCL11 A on cell cycle,proliferation,apoptosis of cervical cancer cells.Methods: Designed and synthesized the siRNA targeted BCL11 A gene,transfect the siRNA into cervical cell C33 A to interfere the expression of BCL11 A,detected the variation of cell cycle,cell proliferation and cell apoptosis via flow cytometry and CCK-8.Results: The expression of BCL11 A were distinctive in different types of cervical cancer cell lines;siRNA1 was the most adequate sequence to silence the expression of BCL11 A in C33 A cells;compared with blank control group(6.56 ± 0.59%)and negative control group(7.85 ±0.47%),the cell apoptosis was up-regulated in siRNA-BCL11 A group(28.45±4.07%)(P<0.05);no significant difference was found when it comes to cell cycle and cell proliferation.Conclusion: BCL11 A expression levels may associated with the type of cervical cancer cell lines;interference of BCL11 A expression in C33 A cells promoted the cell apoptosis but had no impact on cell cycle or cell proliferation.
