With the study of intestinal flora,the relationship between the pathogenesis of ucerative colitis.In this study,the effect of Bifidobacterium(MIMBb75)on body weight,hemoglobin concentration,colon lengh,colon injury and intestinal flora in mice with ulcerative colitis(UC).Firstly,acute ulcertive colitis induced by 2%,3% or 5% of DSS was used.The change of general activity weight,hemoglobin concentration,colon lengh and colon injury was detected to evaluat which is the optimum.Secondly,a total of 63 mice was randomly divided into seven groups,each with 9,including control group,DSS group,MRS group,Mesalazine group,1×108MIMBb75 group,1×107MIMBb75 group and 1×106MIMBb75 group,to discuss the effects of MIMBb75 on the intestinal microflora of ulcerative colitis induced by DSS.1.The results of DSS induced acute ulcerative colitis model in mice were revealed that: the symptoms induced by 3% DSS in mice were similar to UC clinical symptoms.It showed that 3% DSS induced acute ulcerative colitis seems more appropriate.2.Though the study of the effects of MIMBb75 on the general activity of mice,the change of the weight,the colon lengh and hemoglobin concentration,induced by DSS showed that MIMBb75 can effectively low state of health,weight loss and anemia induced by DSS and increase the length of the shortened colon,showed a concentration-response-relationship.3.Though the study of the effects of MIMBb75 on the colon injury in mice induced by DSS showed that MIMBb75 can effectively improve colon mucosa damage index.4.Take the colon contents out under integral asepsis condition and total genomic DNA of bacteria was extracted.The changes of intestinal flora were assayed by polymerase chain reaction denaturing gradient gel electrophoresis(PCR-DGGE)and Cloning and sequencing of the bands.And the the diversity and similarity of each test group were analyzed by cluster analysis.The results showed that: The PCR-DGGE band numbers in three Bifidobacterum MIMBb75 treatment groups were all higher than in the control group and DSS group,and especially,in high-dose Bifidobacterium group the number of bands was significantly increased(P<0.05).The similarity coefficient of three Bifidobacterum MIMBb75 treatment groups was high.Bacteroides sp and Citrobacter sp were commonly detected and [Clostridum] leptum strain and [Clostridium] lactatifermentans were specific.MIMBb75 can reduce the inflammation of UC.These results suggested that Bifidobacterum MIMBb75 can significantly improve the diversity of the microflora and inhibit the development of colitis induced by DSS.
