Objective:The objective of this paper was to investigate the expression difference and clinical significance of Proline-rich tyrosine kinase 2(Pyk2)and phospho-protein kinase B(p-AKT)in tongue squamous cell carcinoma(TSCC)and the adjacent non-tumour tissues.Methods:45 cases of TSCC tissues in our hospital from January 2012 to May 2016 were selected as the research objects,and 30 cases of the adjacent non-tumour tissues were matched.Immunohistochemistry was used to detect the expression of Pyk2 and p-AKT in TSCC and the adjacent non-tumour tissues,so as to study their relevance to clinicopathological parameters of TSCC such as sex,age,tumor diameter,histopathological differentiation types,clinical stage and regional lymph node metastasis by SPSS19.0 statistical software.Results:(1)Pyk2 protein and p-AKT protein expression level were significantly higher in the TSCC tissues,and the expression level of Pyk2 and p-AKT in the adjacent non-tumour tissues were in contrast(P<0.05).The positive rate of Pyk2 protein was 57.78% ?6.67%(?2=17.937,P=0.000),and the positive rate of p-AKT protein was 55.56%?16.67%(?2=9.780,P=0.002)in the TSCC tissues and the adjacent non-tumour tissues.The difference of two groups has obvious statistical significance.(2)The positive rate of Pyk2 in pathological differentiation types was detected.The high differentiation was about 46.87%,the middle and low differentiation is about 84.62%(P < 0.05).In TNM clinical stage,the positive rate of ?-? staging was about 41.94%,?-? staging was about 92.85%(P < 0.05).The positive rate with regional lymph node metastasis group was about 91.67%,and without regional lymph node metastasis group was about 45.45%(P < 0.05).The expression of Pyk2 protein was associated with histopathological differentiation types,regional lymph node metastasis and TNM clinical stage,which is not associated with age,gender nor tumor diameter.(3)The positive rate of p-AKT in pathological differentiation types was detected.The high differentiation was about 43.75%,the middle and low differentiation is about 84.62%(P < 0.05).The difference has statistical significance.The p-AKT expression was only related to histopathological differentiation types,and had no obvious correlation with other clinicopathological parameters.(4)In 45 cases of tongue cancer group,the positive rate of Pyk2 was 26 cases,including p-AKT expressing positive in 16 cases.In 30 cases of the adjacent paracancerous tissues,Pyk2 and p-AKT protein expressing negative was 13 cases.The expression rate of Pyk2 protein and p-AKT protein showed a positive correlation in tongue carcinoma tissues by using statistics(P =0.005,?s=0.412).Conclusion:Pyk2 and p-AKT protein does over-express in the TSCC tissues,but the expression of Pyk2 and p-AKT in the adjacent non-tumour tissues were in contrast.The expression of Pyk2 protein was associated with histopathological differentiation types,regional lymph node metastasis and clinical stage,and the p-AKT expression was only related to histopathological differentiation types.The p-AKT represents the activation state of the P13K/AKT signaling pathway.Pyk2 was the upstream molecule of the pathway.There is a positive correlation between the two proteins,suggesting that Pyk2 may cause or promote the occurrence and development of tongue cancer by participating in the P13K/AKT signal transduction pathway.Pyk2 and p-AKT could be a new target for treatment,and be applied as a new index of monitoring,early screening,diagnosis and prognosis of tongue squamous cell carcinoma.
