Purpose:Most malignant tumors of oral and maxillofacial region belongs to squamous cell carcinoma,it accounts for more than 80% of maxillofacial malignant tumors and most of them are tongue squamous cell carcinoma.In China,tongue squamous cell carcinoma is common in middle-aged and elderly men,and there is early lymph node metastasis and distant metastasis in late stage.There is no strong specific index for the diagnosis of TSCC.In the transcriptional activity,the activation domain required for transcription is completely integrated with the lack of Med19 subunit or the central component of the complex,so the ability of MED to activate transcription is completely inhibited.Studies have shown that high expression of Med19 can be found in a variety of human tumors,and in vitro experiments found that inhibition of MED19 can cause tumor cell line growth inhibition and apoptosis.The experimental study on the expression of MED19 in carcinoma of the tongue,and to study the clinical characteristics of patients with correlation and survival prognosis correlation,to explore the diagnosis and treatment of MED19 in squamous cell carcinoma of the tongue in the future.Methods:We collected 50 TSCC tissues paired with corresponding adjacent non-cancerous tissuesfrom patients who underwent surgery between January 2012 to September 2016 in the Department of Oral and Maxillofacial Surgery,the Affiliated Yantai Yuhuangding Hospital of Qingdao University.The expression of MED19 protein in TSCC and adjacent tissues was detected by immunohistochemistry and Western blotting.In addition,20 paraffin-embedded archived normal oral samples were obtained from our hospital for immunohistochemistry.The expression of MED19 was detected by immunohistochemistry.The relationship between MED19 and age,sex,histological grade,stage and lymph node metastasis were analyzed by statistical method.From January 2012 to June 2014,24 cases of tongue cancer were collected in Yantai Yuhuangding Hospital affiliated to the Qiingdao University.Data were obtained by review or telephone follow-up.Univariate and multivariate COX regression were used to analysethe relationship between MED19 and prognosis of patients,Kaplan-Meierwere used to analyse the relationship between MED19 and patient survival.Results:Immunohistochemical assay showed that MED19 has a high stained brown expression in the nucleus and cytoplasm of TSCC.The positive rate of MED19 was 58%(29/50)in TSCC tissues,the positive rate in adjacent normal tissues was 22%(11/50),and the positive rate in normal oral samples was 20%(4/20)(P< 0.05).The positive rate with lymph node metastasis group was about 88.2%,positive rate without lymph node metastasis group was about 42.4%(P< 0.05).The expression of MED19 protein was associated with regional lymph node metastasis and TNM stage,there was no significant difference in the expression of MED19 protein in patients with gender,age,tumor size nor degree of differentiation.Univariate and multivariate COX analysis showed that the expression of MED19 was correlated with the prognosis of the patients.The survival analysis showed that the survival rate of MED19 high expression group was significantly lower than that of the low expression group.Conclusion:The differential expression of MED19 in tongue cancer tissue and adjacent tissuessuggestedthat MED19 protein expression is associated with squamous cell carcinoma of the tongue.The results of statistical analysis showed that the expression of MED19 protein in tongue cancer patients corelated with lymph node metastasis and TNM stage(P<0.05);but with age,gender and degree of differentiation wereindependent,(P>0.05);the expression of MED19 was correlated with the prognosis of patients,and the overall survival rate of MED19 high expression groupwas significantly lower than that of MED19 low expression group.
