Objective To explore the possibilities of 280 nm UV(ultroviolet)light applied to treating flap ischemic reperfusion injury,then observe the reuslts of flap surviving and talk about the related or possible mechanisms.Methods Fourty Wistar rats were used,they are growing up in the same situation and in the same period;they were divided into four groups by random and 10 rats each,they are group 280nm?group 320nm?blank group and substitute group,the setting of substitute group is just in case for the accident death or another reasons misleading the experiment results.All rats were feeding in the animal laboratory one week,and fasting one day before the peration,abdominal cavity anesthesia by using 10% chloral hydrate,complete the modle of ischemic reperfusion injury on the one side of rat abdomen,and then using 280 nm and 320 nm LED UV irradiate the ischemic reperfusion flap one time about 4 minutes,then feeding by single cage in normal situation and using the UV LED irradiate the relevant rats 4 minutes every two times one day,sustaining a week.The blank group feed in single cage and in normal situation without doing any other things.Observe the flaps of every group rats,and taking photos in time.Especially when we foud the bite of the flap and will affect the experiment results,then we should do the relevant experiment again,so that can substitute the experiment to keep it going on in normal condition.After one week,calculate the square of flap survival by using the method of coordinate stichers,and then kill the rats.Detecting vascular endothelial growth factor(VEGF)by immunohistochemical staining and at the same time by obseving the apoptosis cell by TUNEL test,then get the experiment results by statistical method between the groups.Results After 7 days,the flap survival rate of experiment group was(62.38±8.64)%,higher than the control group(54.49±4.48)% and the blank group(39.61±4.22)%(P<0.05).The density of micrangium and VEGF of experiment group was(91.39±9.29)%and obviously higher than the other two groups of control group was(70.46±4.02)% and blank group was(34.97±3.08)%(P<0.05).By TUNEL test,the amount of apoptosis cell of experiment group was(53.06±5.64),less than the amount of control group(64.72±3.73).Conclusion The local application of 280 nm LED UV can promote the flap survival rate by obviously increase the expression of VEGF,then to generate much more new blood vesselsand.At the same time the apoptosis rate of flap cells is increasing by the midwave UV irradiating locally,however apoptosis rate of the group of less shorter UV is not serious,it possibly affected by its own features of penetrating power and the absorbing condition of the skin.
