Objective We aim to study the expression of PTN in pancreatic cancer cell line Miapaca-2 and Capan-1,and detect the expression quantity of PTN mRNA and protein.This previous study provides scientific basis for later establishing the pancreatic cancer mouse model in situ,by means of disturbing the expression of PTN in pancreatic cancer mouse model.Thereby verifying wether PTN binding to its receptor activitting the relevant signaling pathway in the perineural invasion of pancreatic cancer.At the same time,PTN could be prefered as a potencial drug target and we can interferer its signaling pathway or synthesize inhibitors of PTN in vitro.In order to achieve the purpose of down-regulating PTN or entirely inhibition,thereby arresting tumor growth and reduce the perineural invation and provide scientific basis for finding a new way of early clinical diagnosis or treatment in pancreatic cancer.Methods Mia PaCa-2 and Capan-1 cells were cultured in the DMEM and 1640 culture medium.We measured PTN mRNA and protein expression in pancreatic cancer cell with Q-PCR and western-blot assay.We also detect the expression location of PTN in pancreatic cancer cell with immunofluorescence assay.Results Q-PCR results showed that,The expression of PTN mRNA was signifycantly increased in MIA PaCa-2 cell line,and compared with Capan-1,the expression of PTN m RNA in MIA PaCa-2 is 1.193 times higher than that in Capan-1,showing higher expression.Gel imaging system showed that PTN protein molecular weight is about 19 KDa,with GAPDH who has 36 KDa protein.Then we analyzed the gray value.The results showed that PTN protein both expressed in MIA PaCa-2 and Capan-1 cells,and the expression of PTN protein in MIA Pa Ca-2 cell line was more obvious than that in Capan-1.Immunofluorescence results showed that the positive expression of PTN was brown granules,mainly located in cytoplasm and nucleus.Then we analyzed the correlation of Q-PCR,Western blotting and immunefluore-scence.The results showed that the expression of PTN in m RNA and protein level were consistent in pancreatic cancer cells.Conclusion Our findings suggest that PTN mRNA and PTN protein exhibiting high expression in both Mia paca-2 and Capan-1 cells and the positive material both located in cytoplasm and in the nuclei of pancreatic cancer cell.Mia PaCa-2 cell line could be a good tool for further research of establishing pancreatic cancer mouse mo-del in situ as a result.These results may also provide a basis for investigating whether PTN combined with its receptor activitting the relevant signaling pathway in the perineural invasion of pancreatic cancer.
