Objective: To observe the effect of the traditional Chinese medicine Modified Zhujing Formula on the expression of hypoxia-inducible factor-1 alpha(HIF-1?)and threonine protein kinase B(PKB/AKT)in choroidal neovascularization(CNV)induced by krypton laser and to explore the mechanism.Methods: Eighty-four BN rats(seven-weeks old,male)were randomly divided into four groups,namely normal control group(group A,n=12),model group(group B,n=24),Modified Zhujing Formula treatment group(group C,n=24)and Conbercept treatment group(group D,n=24).The right eyes of group B,C and D underwent fundus krypton laser photocoagulation.Group C was treated with Modified Zhujing Formula for 21 days(10 m L/kg.d),group B was given intragastric administration with equivalent physiological saline and group D was treated by intravitreal injection of a Compaqsepr 21 days after photocoagulation.After modeling the examinations of Fundus color photography,fundus fluorescein angiography,HE staining and immunohistochemistry were performed 28 days,35 days and 42 days after model establishment respectivly to observe the protein expression at different time points.Results: FFA results showed that there is no fluorescence leakage in group A and the fluorescence leakage area in group B is larger than group C and group D at each time point.While that in group C is larger than group D.HE staining.At each time point,the retina and choroid structure of A group were complete and continuous,and B,C and D three groups were found to be damaged by retina and choroid structure.Immunohistochemical showed that there is no significant difference between group B and group C on the expression of HIF-1?(P=0.17?0.05)and AKT(P=0.106?0.05)twenty-eight days after laser photocoagulation.35 d and 42 d after model establishment,the expression of HIF-1?and AKT in group B were significantly higher than that of group C(P<0.01),and the expression of HIF-1? and AKT in group C were significantly higher than that of group D(P<0.05).The expression of HIF-1? and AKT were obviously inhibited in the Conbercept treatment group.Conclusion: Modified Zhujing Formula can prevent the growth of choroidal neovascularization(CNV)induced by krypton laser via inhibit the expression of HIF-1? and AKT.
