Objective: We through the preparation of chronic mountain sickness model in rats,study the changing of the exprsession of FPN?TMPRSS6 and HJV in CMS rats,to reveal the changing of factors related to iron metabolism,to enrichthe content of CMS iron metabolism.Methods: We choosed fifty male heathy SD rats,randomly divided into hypoxia groups and normoxia groups,normoxia groups were respectively fed in Xining(altitude 2260m)seven day,fifteen day,thirty day,every group had eight rats,hypoxia groups were respectively fed in low pressure oxygen cabin(simulated altitude 5000m),hypoxia seven day had eight rats,hypoxia fifteen day had ninerats,hypoxia thirty day had nine rats.In keeping seven day,fifteen day,thirty day,we used peripheral blood to measure blood routine examination,the levels of rats liver FPN?liver mHJV?liver TMPRSS6?muscle sHJV mRNA were detected by qRT-PCR,the protein levels of rats liver mHJV?liver TMPRSS6?muscle sHJV mRNA were measured by Western blotting,the liver FPN protein levels were measured by IHC.Results:(1)The levels of FPN mRNA expression of rat liver in hypoxia seven day,hypoxia fifteen day and hypoxia thirty day were higher than that in normoxia control groups(all P<0.01).Among hypoxia group,the levels of FPN mRNA expression in hypoxia fifteen day and hypoxia thirty day were higher than that in hypoxia seven day(both P<0.01).(2)The levels of liver FPN protein expression of in hypoxia seven day and hypoxia thirty day were higher than that in normoxia control groups(both P<0.01),and the levels of FPN protein expression in hypoxia fifteen day was higher than that in normoxia fifteen day(P<0.05).The FPN protein levels in hypoxia fifteen day and hypoxia thirty day were higher than that in hypoxia seven day(both P<0.01),and the FPN protein levels in hypoxia thirty day were higher than that in hypoxia fifteen day(P<0.05).(3)The levels of mHJV mRNA expression of rat liver in hypoxia fifteen day and hypoxia thirty day were lower than that in corresponding normoxia control groups(P<0.05,P<0.01).The mHJV mRNA levels in hypoxia thirty day was lower than that in hypoxia fifteen day(P<0.05).(4)The sHJV mRNA level of rat muscle in hypoxia seven day,hypoxia fifteen day and hypoxia thirty day were higher than that in corresponding normoxia control groups(all P<0.01).(5)The level of liver TMPRSS6 mRNA expression,hypoxia seven day,fifteen day,hypoxia thirty day were higher than their normoxia control groups(all P<0.01);hypoxia thirty day were higher than hypoxia seven day and hypoxia fifteen day(both P<0.01).(6)The contnt of liver mHJV protein,hypoxia seven day,fifteen day,thirty day were lower than the control group(all P<0.01),with the extension of hypoxia time,mHJV content gradually reduced.(7)The level of muscle sHJV protein expression,hypoxia seven day,fifteen day,thirty day were higher than the control group(all P<0.01),and by the hypoxia time prolonged,the sHJV protein content incrementally increase.(8)The rats liver TMPRSS6 protein content,hypoxia seven day,fifteen day,hypoxia thirty day were higher than their normoxia control groups(all P<0.01),with the extension of hypoxia time,TMPRSS6 content were increased.Conclusion:Under the condition of high altitude hypobaric hypoxia,the mRNA and protein expression levels of FPN,sHJV,TMPRSS6 were increased,and gradually increased with the extension of hypoxia time.mHJV mRNA and protein expression level decreased,and with the extension of hypoxia time gradually reduced.FPN,mHJV,sHJV and TMPRSS6 involved in the regulation of iron metabolism in CMS and may be as important targets for theregulation of iron metabolism in CMS.
