Abstract/Summary:
Objective: Adipose tissue regulates energy metabolism by expressing and secreting adipokines.The abnormal secretion of adipokine promotes the development of metabolic and cardiovascular disease in obesity.As a novel adipokine,complement-C1q/TNF-related protein 12(CTRP12)belongs to the CTRP family,which plays positive role in metabolic and cardiovascular disease.Previous studies found that the serum level of CTRP12 in obesity was decreased.Further mechanism study found that,CTRP12 improves glucose metabolism by inhibiting fat tissue inflammatory response.The latest data show that nearly 40 percent of men and 30 percent of women are overweight.Therefore,the aim of the present study was to measure the serum CTRP12 level in overweight and obesity individuals and analyze their correlations with biochemical index,to explore the clinical significance of CTRP12 in overweight population.Methods: This study was conducted in Health Examination Center of Hebei General Hospital from March to June,2016.Eventually,a total of 282 individuals were enrolled,including the control group(CON,81 subjects),overweight group(OW,93 subjects)and obesity group(OB,108 subjects).Blood glucose,blood lipids and other parameters were analysed by automatic biochemical analyzer.Serum insulin was detected by electrochemical luminescence method.Glycosylated hemoglobin(Hb A1c)was detected by high performance liquid chromatography.CTRP12,APN and Resistin levels were detected by enzyme-linked immunsorbent assay(ELISA)kit.The homeostasis model assessment of insulin resistance(HOMA-IR)was used to evaluate IR.All statistical analyses were performed using SPSS 21.0 software.Results: 1 Comparison of the general c linical and biochemi cal data biochemical data1.1 The general clinical data of participantsCompared with the CON group,BMI,WC and WHR were increased in OW and OB group(P<0.001).And the levels of SBP and DBP in OB group were higher than that in CON group(P<0.001).Compared with OB group,the levels of SBP,DBP,BMI,WC and WHR decreased significantly in OW group(P<0.05).1.2 Comparison of biochemical indexCompared with CON group,the levels of FBG,2h PBG,FINS,2h INS,Hb A1 c,TG,LDL-C in OW and OB group were higher,while the levels of HDL-C and Apo A1 were lower(P<0.05).The level of TC in OB group was much higher than CON group.Compared with OB group,the levels of FBG,2h PBG,FINS,2h INS,Hb A1 c and TG in OW group decreased,while the levels of HDL-C and Apo A1 increased(P<0.05).1.3 Comparison of insulin sensitivity indexCompared with CON group,HOMA-IR index in OW and OB group increased significantly(P<0.001).HOMA-IR index in OW group decreased compared with OB group(P<0.001).2 Comparison of adipokines levels 2.1 The levels of serum adipokinesCompared with CON group,the levels of CTRP12(P=0.03,P=0.001)and APN(P=0.001,P<0.001)were lower in OW and OB groups.While the level of Resistin was higher(P=0.034,P<0.001).The levels of CTRP12 and APN showed decrease trend(P>0.05),while Resistin level increased significantly in OW compared with OB group(P=0.04).2.2 The levels of serum adipokines according to sexAmong the 282 enrolled individuals,133 were men and 149 were women.The differences between sex groups were compared using nonparametric test analysis.The results showed that there were no significant differences in serum levels of CTRP12,APN and Resistin among sex groups.3 Correlation of serum CTRP12 with biochemical related index 3.1 The correlation of serum CTRP12 level with general clinical andThe correlation of serum CTRP12 with general clinical and biochemical data was conducted by Spearman’s or Pearson’s correlation coefficient.The results showed that serum CTRP12 level was negatively associated with age,BMI,WC,WHR,SBP,FBG,2h PBG,Hb A1 c,HOMA-IR,TC,TG and LDL-C.And was no significantly correlation with FINS,2h INS,HDL-C,Apo A1 level.3.2 The influence factors of serum CTRP12 levelThe influence factors of CTRP12 were analyzed.CTRP12 was considered as the independent factor,and multiple linear stepwise regression analysis was used to detect the influence of BMI,WC,WHR,FBG,Hb A1 c,HOMA-IR and TG on the level of serum CTRP12.The results indicated that WC was the important factor of serum level of CTRP12(P=0.001).Conclusion:1 The level of CTRP12 has decreased significantly in overweight individuals.It showed that CTRP12 maybe an early predictor of obesity.2 Serum CTRP12 was negatively associated with age,BMI,WC,WHR,HOMA-IR,FBG,2h PBG,TC,TG,LDL-C,Hb A1 c and SBP.WC was the important influence factor of CTRP12.
