Objective: Through the establishment of the H22 hepatic cancer mice model,the tumor-bearing mice were treated by Hua Zhuo Jie Du Fu Zheng Prescription and 5Fu as control group.In order to investigate the possible mechanism of anti-tumor by Hua Zhuo Jie Du Fu Zheng Prescription,it is that the indexes were observed,which including the general conditions,the inhibition rate of the tumor,the related immune index,the Fas and PCNA'expression in the tumor tissue.Thus,it provides a theoretical basis on clinical application.Methods: The cleaning grade of 50 healthy male Kunming Mice,were randomly divided into two groups according to weight,10 mice in the blank group?N?,40 mice in the model group.Under the aseptic conditions,by hypodermic inoculation in the right front axilla of mice,we established the H22 hepatic cancer mice model for model group.On the princ iple of randomization,40 of tumor-bearing mice were subdivided into four groups,which including the model group?M?,the TCM group?Z?,the 5Fu group?X?,and the TCM and 5Fu group?Z+X?,each group 10.Each group was given the corresponding drug intervention 12 days in a row after 24 hours.After 12 h of the last dose,the mice were killed by the bone marrow dislocated method.Then,open abdominal cavity and blunt dissection tumor,spleen,thymus,which were weighed respectively.The last,we calculated the tumor inhibiting rates,the thymus index and the spleen index and detected the expression level of Fas and PCNA of tumor tissues which fixed in 10% neutral formaldehyde with the immunohistochemical method.Results:1 The comparison of general condition of the miceCompared with the blank group mice,the tumor-burdened group mice were showed listlessness,unresponsive,fewer activities,without burnis h fur and significantly increased diet intake.The model group performance was more outstanding.Among the treatment groups,Z+X group mice performance was better than the others,the X group' performance was poor than the Z group'.2 The comparison of the inhibition rate of the tumor among the groupsAmong the treatment groups,the latest occurrence time of the tumor was the Z+X group,then next was the X group,the Z group,the earliest was the model group.The Z+X group' inhibitory rate was highest,the second was the X group,and the third was the Z group.The average weight of tumor in the Z+X group was the lowest,which was followed by the X group,the Z group,and the model group.As for the average tumor weight,the three treatment group' were less than the model group',the difference was statistically significant?P<0.05?;The Z+X group' average tumor weight was less than the X group' significantly?P<0.05?;the last,the X group and the Z group,have no difference?P>0.05?.3 The comparison of thymus index in different groupsCompared with the normal control group,all the tumor-burdened group mice' thymus index decreased in different degrees,the difference was statistically significant?P<0.05?;Among the tumor-burdened groups,the thymus index of the Z+X group and the Z group were higher than that of the X group,the difference had statistical significance?P<0.05?;but there was no statistical differences between the Z+X group and the Z group?P>0.05?.The X group and the model group were no significant difference of thymus index?P>0.05?.4 The comparison of spleen index in different groupsCompared with the normal group,all the tumor-burdened group mice' spleen index decreased in different degrees,the difference had statistical significance?P<0.05?;Among the tumor-burdened groups,the spleen index of the Z+X group and the Z group were higher than that of the X group,the difference was statistically significant?P<0.05?;but there was no statistical differences between the Z+X group and the Z group?P>0.05?.The X group and the model group had no significant difference of spleen index?P>0.05?.5 The comparison of cancer pathology by HE technique in different groupsCompared with the model group,the tumor cells by HE technique of the treatment groups appeared nuclear pyknosis in different degrees.Additionally,the variations inc luding Splitting reduc ing,revascularization to reduce,more death of tumor cells,increas ing of the lymphocytes.Among the treatment groups,the most obvious is the Z+X group.6 The comparison of the Fas expression of tumor-tissue in different tumor-burdened groupsAccording to the method of immunohistochemistry,the Fas expression of tumor-tissue of treatment groups increased in different degrees compared with the model group,the difference was statistically significant?P<0.05?;Among the treatment groups,the Z+X group was the highest,and the difference had statistical significance compared with the Z group and the X group?P<0.05?;the Z group and the X group had no significant difference?P>0.05?.7 The comparison of the PCNA expression of tumor-tissue in different tumor-burdened groupsAccording to the method of immunohistochemistry,the PCNA expression of tumor-tissue of treatment groups decreased in different degrees compared with the model group,the difference was statistically significant?P<0.05?;The Z+X group was the lowest among the treatment groups,and the difference had statistical significance compared with the Z group and the X group?P<0.05?;the Z group and the X group had no significant difference?P>0.05?.Conclusion:1 Hua Zhuo Jie Du Fu Zheng Prescription can improve the general condition in H22 tumor-burdened mice,increase the inhibitory rate,reduce the tumor weight.It has played a positive role in inhibiting tumor growing.2 Hua Zhuo Jie Du Fu Zheng Prescription can increase the thymus index and spleen index of the tumor-burdened mice;this showed the granules had trigger effecting for immune function in mice.3 Hua Zhuo Jie Du Fu Zheng Prescription can increase the expression of Fas in H22 tumor-bearing mice;while reduced the expression of PCNA.According to the result,we speculated that promote apoptosis and inhibit the tumor cell growth may play an important role in anti-tumor mechanism in mice.
