Objective: To investigate the expression of Notch-1,Jagged-1 and Hes-1in Notch signaling pathway in rectal carcinoma tissuesues and normal paracarcinoma tissues,and to explore the interaction between Notch signaling pathway and the impact of invasion and metastasis on rectal cancer.Method:Real-Time PCR and Western Blot were used to detect the expression of Notch-1,Jagged-1 and Hes-1 in rectal carcinoma tissues and normal paracarcinoma tissues of 40 rectal cancer patients in Hebei Medical University from Augest to November.Statistical analysis software SPSS21.0 was used to analyze the test results.Results:1 The expression of Notch-1 in rectal carcinoma tissues and normal paracarcinoma tissues was detected by real-time PCR.The results showed that the CT values of Notch-1 in rectal carcinoma tissues and normal paracarcinoma tissues were(0.0006564±0.0001491),t=4.327,P=0.00049,the difference was statistically significant(P<0.05),indicating that the expression level of Notch-1 in rectal carcinoma tissues was obviously high than in normal paracarcinoma tissues.2 The expression of Notch-1 in rectal carcinoma tissues was calculated by stratification of the samples according to the gender,age,lymph node metastasis,TNM stage and differentiation degree.In the group of gender,male(0.003394±0.0003704),female(0.003515±0.0003992),t=0.16,P=0.331(>0.05);In the group of age,over 64(0.003421±0.0003617),under 64(0.003510±0.0003747),t=0.97,P=0.305(> 0.05);In the group of lymph nodes,positive(0.003854±0.0003459),negative(0.002985±0.0003571),t=3.52,P=0.001(P<0.05);In the group of TNM stage,T1-T2(0.002814± 0.0004542),T3-T4(0.003954±0.0005172),t=3.35,P=0.002(<0.05);In the group of differentiation,high-medium(0.003031±0.0004012),low(0.004120±0.0004581),t=3.26,P=0.003(P<0.05).It can be seen,Notch-1 expression in rectal cancer was related to the degree of lymph node metastasis,TNM stage and tumor differentiation.3 Western Blot was used to detect the expression of Jagged-1 in rectal carcinoma tissues and normal paracarcinoma tissues.The results showed that the expression of Jagged-1 in rectal carcinoma tissues was significantly higher than that in normal paracarcinoma tissues(t=25.45,P=0.001(<0.05).The difference was statistically significant.4 Western Blot was used to detect the expression of Hes-1 in rectal carcinoma tissues and normal paracarcinoma tissues.The results showed that the expression of Hes-1 in rectal carcinoma tissues was significantly higher than that in normal paracarcinoma tissues(t=35.64,P=0.0089(<0.05).The difference was statistically significant.5 We used correlation analysis to detect the relationship of expression between Notch-1 and Jagged-1 in rectal cancer.r=0.64,when the calibration level was ?=0.05,r=0.30.The expression of Notch-1 was positively correlated with the expression of Jagged-1 in rectal cancer.6 We used correlation analysis to detect the relationship of expression between Notch-1 and Hes-1 in rectal cancer,r=0.45,when the calibration level was ?=0.05,r=0.30.The expression of Notch-1 was positively correlated with the expression of Hes-1 in rectal cancer.Conclusion:1 In Notch signaling pathway,Notch-1?Jagged-1 and Hes-1 were highly expressed in rectal carcinoma tissues and were significantly higher than those in normal paracarcinoma tissues which may be related to the development of rectal cancer,and may became a potential target for the treatment of rectal cancer.2 The expression of Notch-1 was related to the degree of lymph node metastasis,TNM staging and tumor differentiation,suggested that it might affect the invasion and metastasis of rectal cancer.The detection of Notch-1has a certain reference for the early diagnosis and prognosis of rectal cancer value.3 There was a positive correlation between Notch-1 and Jagged-1 and Hes-1 expression in rectal carcinoma tissues,suggesting that this three may be involved in the regulation of Notch signaling pathway and participate in the occurence and development of rectal cancer.
