Objective: Ovarian cancer is a serious threat to women 's life and health of malignant tumors,the mortality rate in the first place of gynecological malignancies.Because the ovaries deep pelvic,the lack of a high degree of sensitivity to the diagnosis of ovarian cancer indicators,resulting in 60%-70% of patients diagnosed at advanced stage.Therefore,the diagnosis and prognosis of ovarian cancer have been the focus of gynecological clinical research and hot spots.In recent years,the role of Hedgehog signaling pathway in malignant tumors has become a hotspot.The abnormal activation of Hedgehog signaling pathway is closely related to the occurrence and development of tumor.It is involved in the regulation of the occurrence,development,proliferation and invasion and metastasis of malignant tumor cells.In this study,the expression of the transcription factor Gli1 and its downstream target gene Foxm1 in the ovarian cancer cell line Skov3 were detected by MTT assay,real-time quantitative PCR and Western blot.The expression of Hedgehog signaling agonist-shh polypeptide was detected to explore the effect of Gli1 and Foxm1 on the proliferation of ovarian cancer cell line Skov3 and its effect on the proliferation of ovarian cancer cell line Skov3 in Hedgehog signaling pathway,and to provide a new theoretical basis for the early diagnosis and treatment of ovarian cancer.Methods:1 Culture of ovarian cancer Skov3 cell line.2(1)The experimental group: adding Hedgehog signal pathway agonist Shh(Human Sonic Hedgehog)polypeptide(200ng / ml);(2)control group:did not add Hedgehog signaling pathway agonist Shh polypeptide.3 The proliferation of ovarian cancer Skov3 cells were detected by MTT assay at 24 h,48h and 72 h.4 The expression of Gli1 mRNA and Foxm1 mRNA in 24 hours,48 h and 72 h of ovarian cancer in each group were detected by real-time fluorescence quantitative PCR.5 Western blot was used to detect the expression of Gli1 and Foxm1 protein in 24 h,48h and 72 h of ovarian cancer in Skov3 cells.6 All data were analyzed by SPSS21.0 statistical software.All data were normal distribution,expressed as mean ± standard deviation((?)±s),the two groups were compared with the t test,P <0.05 was statistically significant.Results:1 MTT assay was used to detect the proliferation of the cells: 24,48,72 hours,the proliferation rate of the experimental group was significantly higher than that of the control group(P <0.05).2 PCR: the expression of Gli1 mRNA and Foxm1 mRNA in the experimental group at 24 h,48 h and 72 h were significantly higher than those in the control group There was significant difference(P <0.05).3 Western blot: The expression of Gli1 and Foxm1 protein in the two groups at 24 h,48h and 72 h were significantly higher than those in the control group at 24 h,48h and 72h(P <0.05).Conclusion:1 Hedgehog signaling pathway agonist Shh polypeptide can increase pathway activity(Gli activity),promote ovarian cancer cell growth.2 Hedgehog signaling pathway,through the expression of high-profile Foxm1 to promote ovarian cancer cell growth.
