| Objective: Oral squamous cell carcinoma(SCC)is one of the most deadly cancers.The maxillofacial defects and dysfunction caused by surgical treatment seriously affect the quality of life of patients.Despite the combination of radiotherapy and / or chemotherapy and other therapies,the 5year survival rate for all patients with SCC is around 50%,and the local recurrence rate was very high[1-2].In order to preserve the integrity of tissues and organs and improve the quality of life of patients,it is urgent to find a comprehensive and effective method.Now there are more and more traditional Chinese medicine have achieved good results for the treatment of malignant tumors,and this trend has increased unabated.In the present study,we analyzed the curative effect and anti-tumor mechanism of Ganluqingliangyin on 7,12-dimethyl benzanthracene(DMBA)-induced oral carcinogenesis in the hamster cheek pouch model,to provide an experimental basis for future experiment and clinical use.Methods: Syrian golden hamsters(6-8 weeks old,100 g in weight,male and female)were randomly divided into five groups,10 for the blank control group,without any treatment.The right cheek pouch of the remaining hamsters was injected with 0.3% DMBA in mineral oil two times a week for12 weeks.Sixty tumor model were then randomly divided into three treatment groups and modle control group(15 in each group).Low dose group,middle dose group and high dose group were fed with 7.5,15,30 g/kg Ganluqingliangyin seven times per week for 4 weeks,modle control group without any treatment.At the 17 th week,all the hamsters were sacrificed and cheek pouch tissue was harvested.After recording the number and volume of visible tumors,the tissues were observed by microscope after HE staining and using immunohistochemical method to detect the BCL-2,CD86 expression in theorganizations.All data using SPSS 13 statistical software,P<0.05 has statistical significance.Results:1 Twelve-weeks of DMBA followed by a 4-week administration of Ganluqingliangyin,high dose of Ganluqingliangyin significantly decreased the incidence from 91.5% to 69.6%(P<0.01),numbers from 2.43±1.55 to1.16±1.02(P< 0.01)and size of oral visible tumor.2 Histopathological observation showed that the number of squamous cell carcinoma in treatment groups was significantly lower than that of the modle group(P <0.05).3 Compared with the modle group,the blood vessel density of the lesions was decreased in each treatment group,especially in the middle and high dose groups(P <0.05).4 Compared with the modle group,the expression level of BCL-2 was decreased in each treatment group,especially in the high dose group(P<0.05).5 The expression of costimulatory molecules CD86 by immunohistochemical detection is higher in each treated group than that in modle group,especially in the high dose group(P<0.05).Conclusions:1 Ganluqingliangyin can inhibit oral buccal mucosa squamous cell carcinoma in golden hamsters,ways in which high doses suppressor effect is best.2 Ganluqingliangyin can inhibit golden hamster buccal carcinoma development via reduction of angiogenesis.3 Ganluqingliangyin may be to promote the apoptosis of buccal carcinoma cell by inhibiting the expression of BCL-2 in tumor tissues.4 Up regulation of the expression of CD86 in immune cells and the improvement of the function of tumor surveillance may be one of the mechanisms of the anti-tumor effect of Ganluqingliangyin. |
