Objective: Oral squamous cell carcinoma(OSCC)is one of the most common malignant tumors in oral,with a high relapse rate and incidence of lymph node metastasis and poor prognosis,and poses potential serious threats to human health.Ganluqingliangyin is composed of ten kinds of Chinese herbal medicine,including Loquat Leaf,Prepared Radix Rehmanniae,Radix Asparagi,Fructus aurantii,Artemisiae Scopariae,Radix Rehmanniae,Ophiopogon japonicas,Dendrobium,liquorice and Radix Scutellariae,and a variety of its constituents have anti-tumor effect.It can inhibit cancer cell proliferation and induce apoptosis of cancer cells,which is expected to become a new drug for the treatment of oral squamous cell carcinoma.Methods: Syrian golden hamsters(6-8 weeks old,100 g in weight,male and female)were randomly divided into five groups,10 for the blank control group,without any treatment.The right cheek pouch of the remaining hamsters was injected with 0.3% DMBA in mineral oil two times a week for12 weeks.Sixty tumor model were then randomly divided into three treatment groups and modle control group(15 in each group).Low dose group,middle dose group and high dose group were fed with 7.5,15,30 g/kg Ganluqingliangyin seven times per week for 4 weeks.At the 17 th week,experimental animal peripheral blood were harvested to detected by flow cytometry after Ganluqingliangyin treatment,applying to observe the effect of Ganluqingliangyin on proportion of CD4+CD25+Foxp3+Tregs and CD80+CD86+ DC in peripheral blood.ELISA was used to evaluate the expression of IFN-??IL-2?IL-4?IL-6?IL-10 and IL-12 in serum.Results:1 After treated with Ganluqingliangyin,low,middle and high dose group of hamster CD4+CD25+Foxp3+Tregs cells in peripheral blood were 1.55 +0.04%,1.20 + 0.03%,0.68 + 0.02%;the proportion of CD80+CD86+DC cells was 0.96 + 0.07%,1.36 + 0.12%,1.70 + 0.14%.results show that with the increase of the concentration of the nectar of cool drink.The proportion of CD4+CD25+Foxp3+Tregs cells decreased gradually,CD80+CD86+ The proportion of DC cells increased gradually(P<0.01).2 After the treatment,the expression level of IFN-,IL-2 and IL-12 in peripheral blood serum of low,middle and high dose groups of golden hamsters were increased,and the expression levels of IL-4,IL-6 and IL-10 were decreased,and there was significant difference among groups.Conclusions:1 Ganluqingliangyin can reduce nectar CD4+CD25+Foxp3+Tregs cells in buccal squamous cell carcinoma of hamster in peripheral blood ratio,increase the expression of CD80+CD86+DC,one of the mechanism of the antitumor effect of mannan is a cool drink that improve the immune antitumor immune response.2 Ganluqingliangyin can reduce the dew of IL-4 squamous cell carcinoma of buccal mucosa of golden hamsters in the peripheral blood IL-6and IL-10 levels,increase IL-2,content of IL-12 and IFN-gamma,suggesting that mannan cool drink may be through the regulation of cytokine expression levels of immune cells to inhibit tumor growth.3 Ganluqingliangyin can change the immune function of the body and improve the immune response of the tumor,which is an effective anti-tumor Chinese medicine prescription.
