Objective: Micro-Plasma Radiofrequency technique(MPR for short)is a new medical method which has been used wildly in clinical works.In plastic department,MPR can benefit us in several clinical treatment such as facial lift、skin regeneration and superficial scar.But there are not studies about the effect of MPR to early hypertrophic scar(HS for short).This study searches the changes of the appearance and the nature and the content of MCP-1 of the hypertrophic scar in rabbit ear model after MPR treatment.This study will supply experimental support for clinical treatment of early HS.Methods: Six New Zealand big-eared white rabbit was departed into different cages and fed for 48 hours in order to let the rabbits adapt the experiment room.Treat rabbit ear HS models by the method of Huiyuan Li.Under the general anesthesia,make use of 8mm diameter circular trephine to form 6 round wound deep to perichondrium along the long axis the ear in the middle abdomen side.There were 72 wounds formed in total at last.Wound scabs appeared 3 days after the HS model form sugery and the epithelium completed near the 7th day.The wounds go into heperplasia peak stage from the 28 th day and this stage will consist for a week.In this stage,the wound scabs had already fall off and the HSes which are red in color and hard in nature can be observed on the surface of the rabbit ears.The 28 th day after surgery,72 HSes were abserved in the ears of 6 rabbits.Averaglely and randomly divided all the 72 HSes into control group and study group.MPR treatment was given to all the 36 HSes in the study group at the 30 th day after surgery.Treatment parameter was a single pass,with 0.6ms and 80 w.The HSes of study group after MPR treatment were applied erythromycin ointment on the surface of the surface of HS,1 times a day,1week.All the 36 HS of the control group were applied erythromycin ointment,1times a day,1 week,without MPR treatment.Cut the HS tissue of control group and control group to be specimens slice for histologic observe at the 60 th day after surgery.And immunohistochemical detection of MCP-1 content in HS was done at the same time.Make use of ImagePro Plus 6.0(Media Cybernetics)image processing software to statistic the area and integral average optical density value of positive cells of each view.T test the integral average optical density value finally using SPSS software.Result:1 Results in naked eye viewThe wound scab of the HS fall off at a average time of 2.4±1.5 days.There were not wound on the surface of the HS after scab falling off.The color of the HS of study group was more lighter and the natue was softer and the height was lower.2 Histological resultsThe dermis of the study group was thinner and there were more fibroblasts hyperplasia and collagen fiber hyperplasia and disordered arrangement compared with the control group.There were not necrotic tissue at the surface of the HS of study group.The fibroblasts hyperplasia was week and the collagen fiber was in line.3 Content of MCP-1The MCP-1 OD of the study group is 0.0175±0.016 and the MCP-1 OD of the control group is 0.054±0.038.The P is 0.00.Conclusion:1 MPR has preventive effect on HS of rabbit ear and MPR is safe without side effects.2 The MPR inhibits the synthesis of MCP-1 to reduce the abnormal fibroblasts proliferation and to inhibit scar hyperplasia.
