Objectives:Effects of different concentrations of extract of Cassia twig heart on succinate dehydrogenase of hypoxic myocardial cells(Succinatedehydrogenase,SDH),carnitine acyltransferase 1(Carnitine Palmitoyltransferase-1,CPT-1)activity and ATP(Adenosine Triphosphate,ATP)the content of and explore the mechanisms of impaired energy metabolism of myocardial cells And provide evidence for correlation between energy metabolism of traditional Chinese medicine in the treatment of heart disease.Methods:Using H9C2 cardiomyocytes in rats were randomly divided into normal control group,model,drug intervention groups,normal control full training in a normal environment.Cells placed in a hypoxic environment,establish model,drug intervention group be different ethanol concentration(40%,60%,80%)of alcohol extract of Cassia twig heart cells to intervene.Observe the changes in myocardial cells,to detect the cardiomyocytes SDH enzyme activity and ATP content,CPT-1.Results:(1)the comparison of SDH: model group compared with normal group,the activity of SDH decrease significantly statistical difference(P < 0.01);Group compared with model group,40% alcohol fetch the SDH activity increases,but no statistical significance(P >0.05),60% alcohol fetch group activities of SDH was statistically difference(P < 0.05),80%alcohol group activities of SDH has statistically significant difference(P < 0.01),with 80%drug curative effect is more significant.(2)the comparison of CPT-1: model group compared with normal group,CPT activity-1 significantly and statistically significant(P < 0.01);Fetch group of 40% alcohol,60%alcohol fetch group compared with model group,CPT-1 was increased,but had no statistical difference(P > 0.05),while 80% alcohol fetch group of CPT-1 rise obviously,statistically significant(P < 0.05).(3)the comparison of ATP: model group compared with normal group,the ATP content decreased statistically difference(P < 0.01);Compared with model group,40% alcohol increases the amount of ATP content group but no statistical difference(P > 0.05),while 60%alcohol fetch group ATP content and statistically significant(P < 0.05),80% alcohol increases the amount of ATP fetch group had statistically significant difference(P < 0.01),with 80%drug curative effect is more significant.Conclusion:Cassia twig sank yangxin alcohol extract can effectively improve the anoxic myocardial cell energy metabolism,the role of approaches may be by adjusting the energy metabolism pathways of succinate dehydrogenase,carnitine fatty acyl transferase activity of enzymes such as 1,make damaged cardiac muscle cells to produce ATP content increases,thus improving the energy metabolism of myocardial cells.Play a role of ingredient is given priority to with 80% ethanol extract.
