Objective: This study was designed to explore the effects of TongNaoWan on the alterations of blood lipid metabolism,AI,lipid oxidation products and levels of vascular endothelial growth factor(VEGF)in brain tissue in experimental model of cerebral atherosclerosis,and it will provide an experimental basis for its clinical application.Methods: A total of 80 Wistar rats was choosed as experimental animals,of which 10 rats were selected as blank group randomly,and remaining others 70 rats were selected as model group.After model were made secuccessfully,a total of 30 eligiable Wistar rats was assigned to next step.The 30 eligiable Wistar rats were divided into three groups randomly,such asmodel group,high and low dosage of TongNaoWan treatment group,10 rats in each group.High and low dosage of TongNaoWan treatment group were given 2.7g/kg.d and 0.9g/kg.d respectively,Model group was given normal saline 3ml/d,and the treatment lasted for 14 weeks.After last treatment,all rats were killed to get serum and brain tissue for further detection.Serum was accumulated to detect serum total cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL),high density lipoprotein(HDL),atherosclerosis index(AI),superoxide dismutase(SOD),malondialdehyde(MDA),glutathione peroxidase(GSH-PX)changes by colorimetric method respectively,brain tissue was collected to detect pathological changes by HE staining,and detect VEGF expression by immunohistochemistry.The difference between each group was analyzed and compared.Results:1.Compared with the model group,the contents of TC,TC,AI,LDL in high and low dosage of TongNaoWan treatment group were significantly decreased,and HDL significantly increased(P<0.05);There was significant difference between the high dose group and the low dose group of TongNao Wan(P<0.05).2.Compared with the model group,the content of serum SOD and GSH-PX in high and low dosage of TongNaoWan treatment group were significantly increased(P<0.05),and the content of serum MDA decreased significantly(P<0.05);There was no significant difference between the high dose group and the low dose group of TongNaoWan(P>0.05).3.Compared with the model group,the expression of VEGF in brain tissue in high and low dosage of TongNaoWan treatment group was significantly increased(P<0.05).There were significant differences between the high and low dose groups(P < 0.05)Conclusion: 1.TongNaoWan could reduce serum TC,TC,LDL content,increase HDL content,and decrease AI index,improve blood lipid metabolism,play a role in anti-atherosclerosis treatment in cerebral atherosclerosis model rats.2.TongNaoWan could reduce serum MDA content,increase SOD,GSH-PX content,alleviate lipid peroxidation injury induced by free radicals,and play a role in anti-atherosclerosis treatment in cerebral atherosclerosis model rats.3.TongNaoWan could increase the expression of VEGF in brain tissue of cerebral atherosclerosis model rats,and play a role in anti-atherosclerosis treatment.
