Objective To explore the association of HLA-A,HLA-B,HLA-DRB1 and HLA-DQ alleles with leprosy in Guizhou Province.Methods Polymerase chain reaction-sequence specific primers(PCR-SSP)was used to genotype HLA-A,HLA-B,HLA-DRB1 and HLA-DQ alleles in 43 patients.23 cases of Healthy adult were enrolled as the control group.Results(1)The allele frequencies of HLA-A*01,03,26,32 were lower in leprosy patients than that in healthy controls,and the allele frequencies of HLA-A*11,24,30,31,33,68 were higher in leprosy patients than that in healthy controls,there were no statistical significance between leprosy patients and the control groups(P>0.05).The allele frequencies of HLA-A*02 was lower in leprosy patients than that in healthy controls(x~2=6.629,P=0.013),the allele frequencies of HLA-A*02 also was lower in LL leprosy patients than that in healthy controls(x~2 =7.935,P=0.0005),suggesting that HLA-A*02may have a negative association with leprosy.(2)The allele frequencies of HLA-B*35,38,40,46,51,52,55 were lower in leprosy patients than that in healthy controls,and the allele frequencies of HLA-B*08,13,15,27,39,56,58 were higher in leprosy patients than that in healthy controls,there were no statistical significance between leprosy patients and the control groups(P>0.05).The allele frequencies of HLA-B*46 were lower in LL leprosy patients than those in healthy controls(x~2 =7.935,P=0.0005),suggesting that HLA-B*46 may have a negative association with LL leprosy.(3)The allele frequencies of HLA-DRB1*01,08,09,12,13,14,16 were lower in leprosy patients than that in healthy controls,and the allele frequencies of HLA-DRB1*03,04,11 were higher in leprosy patients than that in healthy controls,there were no statistical significance between leprosy patients and the control groups(P>0.05).The allele frequencies of HLA-DRB1*15 was higher in leprosy patients than that in healthy controls(x~2 =3.987,P=0.046),the allele frequencies of HLA-DRB1*15 also were higher in LL leprosy patients than those in healthy controls(x~2=4.840,P=0.028),the allele frequencies of HLA-DRB1*16 was lower in LL leprosy patients than that in healthy controls(P=0.019),suggesting that HLA-DRB1*15 may have a positive association with leprosy and that HLA-DRB1*16 may have a negative association with LL leprosy.(4)The allele frequencies of HLA-DQB1*02,04,05,06 were lower in leprosy patients than that in healthy controls,and the allele frequencies of HLA--DQB1*03 was higher in leprosy patients than that in healthy controls,there were no statistical significance between leprosy patients and the control groups(P>0.05).There was no statistical significance between LL leprosy patients and the control groups in The allele frequencies of HLADQB1(P>0.05).There also was no statistical significance between TT leprosy patients and the control groups in The allele frequencies of HLA-DQB1(P>0.05).Conclusion(1)HLA-A*02 may be associated with inhibiting the development of leprosy,that may be protective genes against leprosy patients.HLA-B*46 and HLA-DRB1*16may be protective genes against LL leprosy.(2)The allele frequencies of HLA-DRB1*15 was higher in leprosy patients and LL leprosy patients than that in healthy controls,as HLA-DRB1*15 may be associated with promoting the development of leprosy,HLA-DRB1*15 may be a susceptibility gene in leprosy patients.(3)HLA-DQB1 are not related with leprosy in Guizhou provice.(4)Compared with the research results of correlation between leprosy and HLA in foreign countries,the results of this study are not the same,suggesting that there be a certain degree of particularity on correlation between leprosy and HLA in Guizhou region.
