Study Of Jiawei-Dahuangzhechong-pill’s Effects Based On PDGF And ERKs Signaling Pathways In IHF-Rats

【Objective】To study the effect of Dahuangzhechong-pill combined with Huangqi and Shuihonghuazi Decoction on the expression of PDGF and ERKs in liver tissue of rats with immune hepatic fibrosis,and to provide experimental basis for TCM Yiqi Huayu Tongluo anti hepatic fibrosis.【Materials and Methods】Using the method of intraperitoneal injection with porcine serum induced rat model of immune hepatic fibrosis.6 weeks later,withmaking model,gavaged rats with Jiawei-Dahuangzhechong-pill Decoction,compared with the positive drug Colchicine.There were normal group,model group,Chinese medicine group and positive drug group.Observed the HEstainingin liver tissue of rats by light microscope;biochemical method was used to detect the serum liver function in rats;determination of HYP in rat liver was tested by alkaline hydrolysis;Western blot were used for evaluating the expression of PDGF-BB,PDGFR and ERK1/2 protein in rat liver;RT-PCR was used to elvauate the expression of ERKs m RNA in rat liver.【Results】1.General situation of rats: From the color,stool,behavior,compared with the normal group,the model group rats had worse color;stool color was shallower,texture was looser;Compared with the model group,the treatment group rats’ symptom were improved.2.HE staining of rat liver: In the normal group,the hepatic lobule structure was complete,the liver cells radiated in all directions from the central vein,with no Degeneration or necrosis.there was no abnormal change in the morphology of hepatocytes.In the model group,the structure of hepatic lobule damaged,the morphology of liver cells was deformed,the arrangement was disordered,and the collagen fibers were thickened.Compared with the model group,the liver inflammation and fibrosis degree of each treatment groups were improved.3.Rat liver function: compared with the normal group,ALT AST ALP r-GGT in the model group were increased significantly(P < 0.01),TP and Tbil had the rising trend(P >0.05).Compared with model group,in the traditional Chinese medicine and western medicine group,ALT AST ALP were decreased significantly(P<0.01),TP and Tbil had the declining trend(P >0.05),traditional Chinese medicinegroup r-GGT decreased significantly(P < 0.05),western medicine group expression did not see obvious difference(P >0.05).traditional Chinese medicine and western medicine group expression did not see obvious difference(P >0.05).4.The expression of Hyp in rat liver tissue:Compared with the normal group,the expression of Hyp in the model group was increased(P<0.01);compared with the model group,both the Chinese medicine group and the Western medicine group were decreased(P<0.05).there was no significant difference between the Chinese medicine group and the Western medicine group(P>0.05).5.The expression of PDGFPDGFR and ERK1/2 protein in rat liver tissue :Compared with the normal group,the expression of PDGF PDGFR and ERK1/2was increased in the model group(P<0.01);compared with the model group,both the Chinese medicine group and the Western medicine group were decreased(P<0.01).there was no significant difference between the Chinese medicine group and the Western medicine group(P>0.05).6.The expression of ERKS m RNA in rat liver tissue: Compared with the normal group,the expression of ERKS m RNA in the model group was increased(P<0.01);compared with the model group,both the Chinese medicine group and the Western medicine group were decreased(P<0.01).there was no significant difference between the Chinese medicine group and the Western medicine group(P>0.05).【Conclusion】Jiawei Dahuangzhechong pills could improve liver fibrosis,protect liver function,and reduce the expression of PDGF-BB,PDGFR and ERKs protein in liver tissue,suggesting the mechanism that Jiawei Dahuangzhechong pill combined with Huangqi and Shuihonghuazi Decoction treats hepatic fibrosis may be related to the inhibition of the proliferation and activation of HSC.This experiment provided some experimental data supporttointerpret the treatment effect of Dahuangzhechong pill combined with Huangqi and Shuihonghuazi Decoction on immune hepatic fibrosis,and provided experimental basis for clinical rational application of Jiawei Dahuangzhechong pill in the treatment of related diseases.
