Objective : To explore the expression and clinical significance of Suppressor of Cytokine Signaling Protein 3(SOCS3)and interleukin 6(IL-6)in ovarian carcinoma,to investigate the effects of IL-6 of different concentrations on cell proliferation and SOCS1 and SOCS3 expressions in human SKOV3 ovarian cancer.Methods:(1)The expression of SOCS3 and IL-6 in 105 cases of ovarian carcinoma,40 cases of benign ovarian tumors and 30 cases of nomal ovarian tissues were explored by immunohistochemistry,and analyzed their relationship with clinicopathological features by relevant statistical methods.(2)The different concentrations of IL-6 were acted on the human SKOV3,then the cell proliferation,and the protein and the mRNA expressions of SOCS1 and SOCS3 were explored by the methods of Cell Counting Kit-8(CCK-8)? reverse transcription polymerase chain reaction(RT-PCR)and Western Blot.Results :(1)The positive rates of expression of SOCS3 in ovarian carcinima was significantly higher than those of benign ovarian tumors and normal ovarian tissues(X2=57.672,P<0.01).At the same time,the positive rates of expression of IL-6 in ovarian carcinima was significantly higher than those of benign ovarian tumors and normal ovarian tissues(X2=44.95,P<0.01).The overexpression of SOCS3 in ovarian carcinima was associatied with the histological classification and ascites(P<0.05),in addition,The positive rates of expression of SOCS3 in high-level ovarian carcinoma with ascites was significantly higher than those of low-level ovarian carcinoma without ascites(P<0.05).The overexpression of IL-6 in ovarian carcinima was associatied with the histological classification,FIGO clinical stage and ascites(P<0.05),in addition,the positive rates of expression of IL-6 in late high-level ovarian carcinoma with ascites were significantly higher than those of early low-level ovarian carcinoma without ascites(P<0.05).There were positive correlations between the expression of SOCS3 and IL-6 by the Spearman's correlation analysis(r=0.574,P<0.05).(2)The different concentrations of IL-6 effected on the human SKOV3,which were dependent on the concentration of and promoted proliferation of the human SKOV3(F=3531.369,P<0.01).The different concentrations of IL-6 effected on the human SKOV3,which were dependent on the concentration of and the mRNA and protein relativeexpressions of SOCS1 and SOCS3 of the human SKOV3.(F=18.832?261.112?6.096?58.922,P<0.01).Conclusion :(1)The SOCS3 was higher expressed in ovarian carcinoma,and the overexpression of SOCS3 in ovarian carcinima were associatied with the histologicall classification and ascites but not Histologic type,Lymph node metastasis,FIGO clinical stage and age.In addition,the positive rates of expression of SOCS3 in high-level ovarian carcinoma with ascites were higher than those of low-level ovarian carcinoma without ascites,which means that the expression of SOCS3 was closely related with the occurrence and development of ovarian cancer.The IL-6 was higher expressed in ovarian carcinoma,and the overexpression of IL-6 in ovarian carcinima was associatied with the histological classification,FIGO clinical stage and ascites but not Histologic type,Lymph node metastasis and age.At the same time,the positive rates of expression of IL-6 in late high-level ovarian carcinoma with ascites were higher than those of early low-level ovarian carcinoma without ascites,which shows that the expression of IL-6 may play a important role in the prognosis of ovarian cancer and contribute to the development of ovarian cancer.(2)The concentrations of IL-6 increasing,decreased the mRNA and protein expressions of SOCS1 and added the mRNA and protein expressions of SOCS3,which indicated that different expressions of SOCS1 and SOCS3 induced by IL-6 can influence the proliferation of human SKOV3 ovarian cancer.
