?Objective?:The research aims at jointly detecting the expression of DCLK1 and Bmi-1 in 74 cases of colorectal cancer,and understanding the relationship DCLK1 and Bmi-1with colorectal cancer.?Method?:Collected 74 cases of colorectal cancer patients with complete pathological and clinical data in Affiliated Hospital of Yan'an University from September 2015 to October2016,which was not offered radical surgery before radiotherapy,chemotherapy and other related treatment,the selected experimental specimens included colorectal cancer and corresponding adjacent normal tissues(Note: the normal tissue is >1cm away from the cancer tissue,and there are no abnormal cells such as cancer cells and adenomatous hyperplasia cells under the microscope).The expression of DCLK1 and Bmi-1 in colorectal cancer was detected by immunohistochemical Max Vision TM method,and statistically analyzed the correlation between them and those relationship with clinicopathological parameters.Using SPSS 20.0 software to statistically analyse all the data.Chi-square test were used to analyze the enumeration data,the Spearman rank correlation analysis were used to analyze the correlation between DCLK1 and Bmi-1,there was a significant difference that the definition of P is less than 0.05,it was a significant difference that the definition of P is less than 0.01.?Result?:1.The positive expression rate of DCLK1 protein in colorectal cancer tissues(68.9%)was significantly higher than that in adjacent normal tissues(27.0%),the yellow or brown yellow staining of immunoreactive products was mainly located in the cytoplasm,which is granular,after chi-square test,x2 =26.016,P<0.01,the difference was significant;with infiltration's depth,lymphatic metastasis and Dukes stage of colorectal cancer,there was significant correlation(P<0.05),and the difference was statistically significant;withgender,age,tumor location,the degree of tumor differentiation and tumor type showed no significant correlation(P>0.05).2.The positive expression rate of Bmi-1 protein in colorectal cancer tissues(62.2%)was significantly higher than that in adjacent normal tissues(23.0%),the yellow,brown yellow or brown staining of immunoreactive products was mainly located in the nucleus,and the less is in the cytoplasm,which is granules,after chi-square test,x2 =23.243,P<0.01,the difference was significant;with the degree of tumor differentiation,infiltration's depth,lymphatic metastasis and Dukes stage of colorectal cancer,there was significant correlation(P<0.05),and the difference was statistically significant;with gender,age,tumor location,tumor type showed no significant correlation(P>0.05).3.The relationship between DCLK1 and Bmi-1 expressed in colorectal cancer tissues showed a positively correlation by Spearman rank correlation analysis(rs=0.259,P=0.026,P<0.05),the difference was statistically significant.?Conclusion?:1.DCLK1 obvioursly expressed in colorectal cancer was significantly correlated with infiltration,lymphatic metastasis and Dukes stage of colorectal cancer.2.Bmi-1 obvioursly expressed in colorectal cancer wass significantly correlated with differentiation,infiltration,lymphatic metastasis and Dukes stage of colorectal cancer.3.There was a positive correlation between the expression of DCLK1 and Bmi-1 in colorectal cancer tissues,suggesting that DCLK1 may indirectly affect the expression of Bmi-1 through some mechanism in colorectal cancer,both of which may be involved in the pathogenesis,progression,invasion and metastasis of colorectal cancer.the expression inhibited of both may prevent the occurrence and development of tumors.The joint detection of DCLK1 and Bmi-1 may provide a new approach for the clinical diagnosis,treatment and prognostic evaluation of colorectal cancer.
