Objective: Curcumin/hyaluronic acid(Cur-HA)nanomicelles were successfully prepared by esterification action.Meanwhile its mechanical properties,biocompatibility and therapy effect on rheumatoid arthritis were tested and evaluated to providing some research experience for its clinical treatment of RA disease.Materials and methods: Cur-HA composite was prepared by esterification action and then self-assembled to nanomicelles in an aqueous solution.The morphology and structure of Cur-HA nanomicelles were characterized by transmission electron microscopy(TEM),dynamic light scattering(DLS)and fourier transform infrared spectroscopy(FTIR).MTT method was used to detect the toxicity of Cur-HA to bovine articular chondrocytes.The rats were randomly divided into control group,RA group,Cur-HA treatment group,HA treatment group,Cur treatment group and saline treatment group.The corresponding drug was injected into ankle joint of the rats once a week,and then the joint swelling rate was detected two weeks later.In addition,TNF-a,IL-l,VEGF,AST,ALT,ALP were detected by ELASA.Moreover,the pathological change of the joint tissue was observed by the histochemical method.More importantly,the friction properties of articular cartilage of achyranthes bidentata under different friction medium were studied by friction and wear testing machine(UMT-2MT)and scanning electron microscopy(SEM).Result and conclusion:1)Characterization of Cur-HA: FTIR shows the characteristic peaks of ester bond in Cur-HA,indicating that HA is successfully connected to Cur.The TEM showed that the Cur-HA nanomicelles were multi layered annular structures,and DLS showed the particle size of Cur-HA nanomicelles less than 164 nm.2)The biological safety evaluation of Cur-HA: in vitro and in vivo experiments showed that Cur-HA nanomicelles had no toxic effect on bovine articular chondrocytes and Wistar rats.3)The clinical evaluation of Cur-HA: After two weeks of treatment,Cur-HA can reduce the joint swelling about 30%(P < 0.05),Cur-HA can inhibit RA rat serum TNF-a,IL-l and VEGF expression(P < 0.05).Histochemical results showed that Cur-HA had good therapeutic effect on RA.4)The lubrication action of Cur-HA:Cur-HA,HA,Cur and NS as the friction medium,the friction coefficient was 0.027±0.006,0.049±0.015,0.085±0.025 and 0.062±0.009.SEM showed that compared with HA group,Cur group and NS group,the Cur-HA group had lower friction marks and smaller friction chips,which indicated that Cur-HA had better lubrication performance.The results of experiments and characterization showed that Cur-HA nanomicelles were successfully synthesized,and Cur-HA nanomicelles had good therapeutic effect on RA.
