BackgroundDepression is a comprehensive variety of factors,and chronic mental illness has a genetic predisposition,its main clinical symptoms of depression or pessimistic remorse,loss of interest,sense of guilty or worthless,somatic symptoms can be characterized by sleep disorders,loss of appetite,decreased activity,or concentration.Depression or pessimistic is the main clinical symptoms of depression,the characteristics of depression is chronic,recurring.It is harmful to people's physical and mental.Depression has become a pandemic of the 21 st century.However,the mechanism of depression is still not clear.In recent year,the role of relationship between the immune system in depression attract more and more people's attention,and gradually become a research hotspot of depression.Epidemiological investigation showed that there is a close correlation between pain and depression,depression incidence of pain and pain people crowd incidence were higher than their single rate,pain and depression of contract rate from 50% to 60% on average.Among them,depression and anxiety rates were around 16 percent in the population,but among chronic pain patients there was a higher rate of about 35 percent.Pain and depression are often associated with common diseases,often overlapping and overlapping,with more impaired function and poorer quality of life.Therefore,it is urgent to treat the common disease with depression.So this article mainly through the bee poison caused by chronic pain,further induce the depression model,to explore the pathogenesis of depression,so as to find antidepressant treatment strategies,thus providing theoretical basis for clinical treatment with antidepressants.Aim:1.To explore the relationship between and cytokins;2.The role of PAK-ERK/p38 MAPK signaling pathway in depression.Method:According to the criteria,86 elderly depression patients and 86 normal elderly patients were selected.The experiment was divided into three groups: normal group,depression group,depression group.All personnel in the early morning,fasting venous blood collection.The expression of MMP-2 and MCP-1 were detected by Western-blot blotting,and the levels of E-selectin and AOPPs were detected by ELISA and spectrophotometer.Adult Spragure-Dawley(SD)was purified and injected with bee venom to establish the model of bee venom induced depression.They were randomly divided into three groups:1.The normal control group(control): the rats were injected with 0.9%normal saline for 50 mL,and injected injected with 0.9%normal saline for 5mLafter 2 hours;2.Bee virus model group(BV+Veh): The rats were injected with 50mL bee venom at the concentration of 4mg/ml.After 2 hours,the rats were injected with5mlsaline;3.The drug treatmen group(BV+SB): The rats were injected with 50mL bee venom at the concentration of 4mg/ml.After 2 hours,the rats were injected with5ml SB203580,the concentration of 1mg/ml.Quickly decapitated,we collected fresh spinal cord tissue,and performed protein extraction.We used Western blot method to detect the phosphorylation of activation of p21 kinase(p21-activated kinase,PAK),p38,extracellular signal regulated kinase(extracellular signal-refulated,kinase,ERK).Result:1.Western blot showed that the expression of MMP-2 and MCP-1 in senile depression group was higher than that of normal control group,and the content of MMP-2 and MCP-1 in serum were significantly lower after oral administration of Pa Rossi Dean for 14 days2.We used ELISA method to detect the concentraction of serum E-selectin and AOPPs.Compared with the normal control group,the serum E-seletin and AOPPs in patients with depression were significantly increased.When treated with oral administration of Pa Rossi Dean,the levels were significantly lower.3.The results showed that the level of PAK phosphorylation in the spinal cord of rats was increased after the injection of bee venom,and the phosphorylation level of PAK was significantly decreased after the treatment with P38 MAPK inhibitor SB2035804.The results of Western blot showed that the level of p38 phosphorylation was increased after the rats were injected with bee venom,suggesting that bee venom could activate p38 activity.When treated with the inhibitor SB203580,the phosphorylation level of p38 was significantly decreased.There was no significant change of total p38 in each group,which indicated that SB203580 treatment had no effect on the expression of total p38.5.protein electrophoresis showed that there was no significant change of the total ERK1/2 in each group,indicating that SB203580 had no effect on the expression of ERK1/2 in the spinal cord.Compared with the normal control group,the level of ERK phosphorylation was significantly increased after the injection of bee venom,suggesting that bee venom induced depression could enhance the ERK signaling pathway.It should be noted that the phosphorylation level of ERK was significantly decreased after SB203580 treatment after the occurrence of depression,immune function related cytokines activation chronic pain activates P38 MAPK signaling pathway and induces depression.
