Zhizi Baipi Decoction is classical prescription,which is used to treat TCM Yang-jaundice syndrome and an eutherapeutic medicine to treat jaundice in modern clinical.In present study,Zhizi Baipi Decoction was selected as the research object.HPLC methods were developed for content determination of active ingredients 、optimizing the extraction technology;Effects of Zhizi Baipi Decoction on rat model with TCM Yang-jaundice syndrome was investigated.Combined with the pharmacodynamics inspection and drug serum chromatogram,the efficacy of the material is explored and mechanism preliminary is discussed,which provide some experimental basis for the clinic.This article are studied from the following four aspects.1、Content determinationTo determinate content of geniposide 、 liquiritin 、 crocin-1 、 jatrorrhizine hydrochloride、 crocin-2、 berberine hydrochloride、 palmatine hydrochloride and glycyrrhizic acid in Zhizi Baipi Decoction by HPLC simultaneously.The eight components were detected effectively and had good resolution.2、Optimization of extraction process(1)To determinate content of geniposide、 liquiritin、 crocin-1、 jatrorrhizine hydrochloride、 crocin-2、 berberine hydrochloride、 palmatine hydrochloride and glycyrrhizic acid in single medicine decoction and mixed medicine decoction of Zhizi Baipi Decoction by HPLC.The contents of total alkaloids and glycyrrhizic acid in the combined decoction were lower than that in the separated decoction.The contents ofcrocin-1 and crocin-2 in the combined decoction were higher than that in the separated decoction.The contents of geniposide and liquiritin in the combined decoction were almost as same as that in the separated decoction.(2)The orthogonal test was employed taking the contents of geniposide 、liquiritin、 crocin-1、berberine hydrochloride and extraction rate as the indexes and the influences on extraction results of soaking time,solvent dosage,extraction times and extraction time were investigated,and were analyzed by multi-index comprehensive scoring method.The optimal extraction technology was as follows: 12 times of ethanol concentration 70% consumption,soak for 60 min,extraction thrice for 1.5 hour at each time.3 、 Therapeutic effect of Zhizi Baipi Decoction on rat model with TCM Yang-jaundice syndrome(1)Rat model with TCM Yang-jaundice syndrome was induced by i.g.α-naphthyl isothiocyanate(APIT),along with the hot and humid environment and high-fat and high-carbohydrate diet.).Several doses of zhizi Baipi Decoction extract were ig administrated to the models,the daily state of rats was observed.The expressions of alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL),bile acid(TBA),complement C3,C4,total superoxide dismutase(T-SOD),malonaldehyde(MDA)were detected and the pathological sections were observed under the light microscope.Compared with model group,Moderate dose of Zhizi Baipi Decoction group and high dose of Zhizi Baipi Decoction group decreased significantly ALT,AST,TBIL,TBA,Complement C3,C4 in blood serum(P<0.01),improve the activity of SOD(P<0.05,P<0.01);Moderate dose of Zhizi Baipi Decoction group decreased the content of MDA(P<0.05),they reduced the pathological injury in liver tissues at different levels.(2)Rat model with TCM Yang-jaundice syndrome was induced by i.g.α-phenyl isothiocyanate(APIT),along with the hot and humid environment and high-fat and high-carbohydrate diet.Several doses of zhizi Baipi Decoction and Its Components extract were ig administrated to the ALT,AST,TBIL,TBA,Complement C3,C4 in blood serum(P<0.05,P<0.01);Gardeniae Fructus-Glycyrrhiza and Gardeniae Fructus groups decreased significantly ALT,AST,TBIL,TBA,Complement C3,C4 in blood serum(P<0.05,P<0.01),Gardeniae Fructus-Glycyrrhiza group decreased the content of MDA(P < 0.05);Cortex Phellodendri-Glycyrrhiza group decreased Complement C3,C4 in blood serum(P<0.05,P<0.01);all groups improved the activity of SOD(P<0.05,P<0.01);they reduced the pathological injury in liver tissues at different levels.4、HPLC spectrum-effect relationship of Medicinal serumThe analysis methods of HPLC chromatograms of Zhizi Baipi Decoction and medicinal serum,which were separated from Rat model with TCM Yang-jaundice syndrome was ig administrated by zhizi Baipi Decoction and Its Components extract,were established.HPLC chromatograms of medicinal serum of Zhizi Baipi Decoction and Its Splited Prescriptions were established.Compared with Zhizi Baipi Decoction,there were 4 chemical compositions absorbed into blood in the medicinal serum,which come from the metabolites.The results of chromatograms showed that2、3、4 peaks came from Fructus Gardeniae and 1 from glycyrrhiza.Zhizi Baipi Decoction and splited prescription 1 groups have positive effects in treatment of TCM Yang-jaundice;metabolites may be material foundations;the mechanism maybe lie in preventing inflammatory response and antagonizing free radical injury.A negative correlation between 2 peak and TBIL,implies that 2 peak may be a medicinal effective component. |
