Abstract/Summary:
Objective To investigate the pathogenesis of premature ovarian failure induced by Tripterygium wilfordii preparation,and to observe the preventive and therapeutic effect of Chinese herbs on the premature ovarian failure.Methods The 12 week old female SD rats were randomly divided into six groups: blank group,model group,Western medicine group,Bushen Tiaochong recipe of prevention group;treatment group and intervention group.Eccept the blank group,the other groups were given tripterygium glycosides tablets 75mg/kg gavage model.Before modeling group treated with Bushen Tiaochong prescription in different doses(36d),and then give Tripterygium wilfordii model(14d);model group given Bushen tiaochon morning dose prevention(36d),afternoon given tripterygium glycosides tablet model(14d);after modeling group first given tripterygium glycosides tablet model(14d),then given different doses of Bushen Tiaochong Recipe treatment(36d).The western medicine control group was given the combination of estrogen and the control group was given the Kuntai Capsule group.Observation of rat behavior change,estrous cycle,ovary,uterus index calculation,using ELISA and radioimmunoassay of serum hormone levels in rats,protein and gene were detected in rat ovarian related factor Immunohis-tochemistry and PCR method.Results 1 Effect on the estrous cycle of rats: compared with model group,each drug group disorder rate had significant difference(P<0.05);compared with the western medicine group,there was statistical significance differences made before model group low dose of Chinese medicine group(P<0.05);model group Chinese medicine low and medium dose group were statistically significant(P<0.05);there was statistical significance after the model group,Chinese medicine group(P<0.05);the difference compared with Kuntai Capsule group,was statistically significant differences before model making group high dose of Chinese medicine group(P<0.05);there was statistically significant difference between the model group in the traditional Chinese medicine,the high dose group(P<0.05);there was statistical significance after the model of traditional Chinese medicine each dose group differences(P<0.05).2 Effect on rats sex hormone levels: compared with the model group,there was statistical significance in each group of serum E2,AMH,FSH,LH level difference(P<0.05);compared with the western medicine group,serum E2,AMH,FSH and LH have statistical significance before modeling group,low dose of Chinese medicine group differences(P<0.05);there was statistical significance in the model group,Chinese medicine low doses group difference(P<0.05);there was significant difference in the model group of Chinese medicine groups(P<0.05);compared with Kuntai Capsule group,serum FSH,LH,E2,AMH showed a significant difference in model group before the Chinese high dose group(P<0.05);there was statistically significant difference between the model group in the traditional Chinese medicine,the high dose group(P<0.05);model group differences of Chinese medicine groups were statistically significant(P<0.05).3 Effect on rats VEGF,b FGF protein and m RNA expression: compared with the model group,the expression of VEGF in each treatment group,b FGF protein and m RNA were significantly increased(P<0.05);compared with the western medicine group,the expression difference was statistically significant before modeling group,low dose of Chinese medicine group,b FGF protein m RNA(P<0.05)had statistical significance;differential expression of model group low dose of Chinese medicine group,VEGF protein m RNA(P<0.05),traditional Chinese medicine,in low dose group b FGF the expression of m RNA protein was statistically significant compared with the western medicine group(P<0.05);after modeling,traditional Chinese medicine,in low dose group VEGF,b FGF protein and m RNA expression was statistically significant(P<0.05).Before making model group and Kuntai Capsule group,high dose of Chinese medicine group VEGF,b FGF,m RNAprotein expression was statistically significant(P<0.05);model group,Chinese medicine high dose group VEGF protein expression difference statistics Chinese medicine(P<0.05);the expression of b FGF protein in the high dose group was statistically significant(P<0.05).The difference of VEGF and b FGF protein expression in each dose group was statistically significant(P<0.05);4 Effect on rats TNF-α,IFN-γ,protein and m RNA expression of: compared with the model group,rats before and after expression in each group of TNF-α,IFN-γ,protein and m RNA decreased significantly(P<0.05);compared with the western medicine group,Chinese medicine group: rats in the high dose group of TNF-α,IFN-γ,m RNA,protein expression had signifycant difference(P<0.05);traditional Chinese medicine group,low dose group after the model had significant difference(P<0.05);compared with Kuntai Capsule group,TNF-α,IFN-γ,m RNA,a protein expression had significant difference before modeling group,high dose of Chinese medicine group(P<0.05);model group in medicine in the high dose group had significant difference(P<0.05),after the model group,Chinese medicine group had significant difference(P<0.05);traditional Chinese medicine in the high dose group had significant difference(P<0.05).Conclusion 1 Tripterygium wilfordii tablets 75mg/kg(14d)can cause premature ovarian failure in rats,estrous cycle disorder,body mass,serum levels of low estrogen and high status.2 Chinese medicine Bushen Tiaochong decoction can treat premature ovarian failure in rats,treatment medicine for Chinese medicine high dose group,dose group,low dose group of Chinese medicine in traditional Chinese medicine;traditional Chinese medicine Bushen Tiaochong Recipe has preventive effect on premature ovarian failure,which before making model group and model group,both play the role of prevention.The effect of preventive medicine in the middle and low dose group was the best.3 Kun Tai capsule can also prevent premature ovarian failure in rats,but the effect of treatment is inferior to the kidney.The same as the kidney regulating and punching,the preventive effect of premature ovarian failure was the most obvious.4 Combined with estrogen can treat premature ovarian failure in rats.The curative effect was equivalent to the middle dose group and the control group in the control group,and the high dose equivalent.5 Chinese medicine Bushen Tiaochong decoction can prevent premature ovarian failure in rats,its mechanism may be some ingredients in traditional Chinese medicine and has a positive effect on gonadotropin increase,promote the recovery of ovarian function,improve ovarian function;the expression of TNF-α,IFN-γ,reduce follicular atresia,slow depletion of follicles;up regulating the expression of VEGF and b FGF the promotion of angiogenesis,vascular cavity expansion,reduce hardening of pipe wall thickness changes of blood rheological properties.6 Tthe establishment of the model group before the establishment of the group,can be given to the kidney in advance of the prevention and control to reduce the degree of damage to the gonads.
