Effect Of Shexiang Wulong Pill On The Expression Of MiR-146a,miR-130 And MiR-223 In The Peripheral Blood Of Patients With RA

Objectives To explore the effect of drug serum of Shexiang Wulong pill on the levels of miR-146 a,miR-130,and miR-223 in PBMCs from Patients with RA.Methods 1 Drug Serum Preparation: Totally 12 SFD SD rats were divided into 3 groups according to the random digital table method.Normal Saline group.The Shexiang Wulong pill group.The TGP(TGP)group.Rats were lavage with 10 m L/kg/d physiological saline,The Shexiang Wulong pill water solvent(200 mg/m L),TGP water solvent(20 mg/m L)for 7 days.We will extract the drug-containing serum by abdominal aorta blood and stored at-80 for later use.2 The study selected? RA patients who was first diagnosed and not be treated with antirheumatic in north China university of technology affiliated hospital rheumatoid immunotherapy clinic from November 2015 to May 2016,aged between 30 and 50 years old.Collecting fresh venous blood in patients with 5 m L,then extracting PBMCs by ultraclean audience.And we will have cell counted after 24 hours culture in condition of 37 and 5% CO2.Then cells were divided into 4 ?groups as follows,control group(medium);serum-free group;drug serum of Shexiang Wulong pill group;4)drug serum of total glucosides of paeony group.After treatment for 48 hours in condition of 37 and 5% CO2,total RNA was extracted ?from PBMCs.Then test the absorbance of the sample RNA in the OD260 nm/OD280 nanometers in order to detect the purity and concentration of the sample RNA.According to the c DNA reverse transcriptional kit(MBI company),the c DNA is synthesized.Take c DNA as a template and U6 sn RNA as the internal reference,the expressions of miRs were detected by real-time quantitative polymerase chain reaction(RT-PCR).The samples of each group were set up with 3 holes,in a total of 40 cycles.The relative expression of each miR was expressed by 2-Delta Ct.Results Serum containing drug prognosis.Compared between blank control group and serum-free group,the level of 3 miR was not statistically significant(P > 0.05).Compared with blank control group and serum-free group,the expression of miR-146 a,miR-130,and miR-223 were decreased in the drug serum groups(P < 0.05).Compared with drug serum of TGP group,the level of miR-223 and miR-146 a were decreased in the experimental group(P < 0.05);the level of miR-130 was not statistically significant(P >0.05).Conclusions 1.Shexiang Wulong pill maybe inhibited inflammation in RA patients by down-regulation the levels of miR-223,miR-130,and miR-146 a.2.The expression level of miRNA-223 and miRNA-146 a in PBMCs of RA patients treated by Shexiang Wulong Pill was lower than that of TGP.
